五味子根茎叶果多糖对D-半乳糖致小鼠学习记忆障碍的影响 被引量:1

Effects of Polysaccharides from root,stem,leaf and fruit of Schisandra chinensis on learning and memory disorder in D-galactose-treated mice

摘要 目的观察五味子根、茎、叶及果多糖对D-半乳糖所致小鼠学习记忆障碍的影响。方法将雄性ICR小鼠随机分为6组,即空白组、模型组、五味子根多糖组、茎多糖组、叶多糖组及果多糖组,分别给予蒸馏水及总糖含量为35mg/kg的根、茎、叶及果粗多糖灌胃,30min后除空白组皮下注射生理盐水外,其他组均皮下注射300mg/kg剂量D-半乳糖,每日1次,持续6周。通过避暗实验和Morris水迷宫实验评价改善学习记忆能力。结果在避暗穿梭实验中,与空白组比较,模型组小鼠避暗潜伏期显著缩短,避暗穿梭次数显著增加(P<0.01);与模型组比较,五味子根、茎和果多糖组小鼠避暗潜伏期显著延长,避暗穿梭次数显著减少(P<0.01)。Morris水迷宫实验中,与空白组比较,模型组小鼠空间探索潜伏期显著延长,目标象限停留时间显著缩短,穿越平台次数显著减少(P<0.01);与模型组比较,五味子根、茎及果多糖组小鼠空间探索潜伏期显著缩短(P<0.05,P<0.01);五味子茎及果多糖组小鼠目标象限停留时间显著延长(P<0.01),根及叶多糖组目标象限停留时间无统计学差异(P>0.05);五味子茎及果多糖组小鼠穿越平台次数显著增加(P<0.01)。结论五味子根、茎及果多糖对D-半乳糖所致鼠学习记忆障碍具有改善作用。 Objective To observe the effects of Schisandra chinensis roots,stems,leaves and fructose polysaccharides on learning and memory disorder in mice induced by D-galactose.Methods Male ICR mice were randomly divided into six groups,namely blank group,model group,Schisandra chinensis root polysaccharide group,stem polysaccharide group,leaf polysaccharide group and fructose polysaccharide group.They were given distilled water and crude polysaccharide from roots,stems,leaves and fruits with total sugar content of 35 mg/kg stomach.After 30 minutes,except for the blank group,which was subcutaneously injected with normal saline,the other groups were subcutaneously injected with D-galactose at a dose of 300 mg/kg,once a day,for a continuous period of time 6 weeks.The ability to improve learning and memory was evaluated by dark avoidance test and Morris water maze test.Results In the dark avoidance shuttle experiment,compared with the blank group,the dark avoidance latency of mice in the model group was significantly shortened(P<0.01),and the number of dark avoidance shuttles was significantly increased(P<0.01).The dark avoidance latency of mice in the group was significantly prolonged(P<0.01),and the number of dark avoidance shuttles was significantly reduced(P<0.01);there was no statistical difference in the number of dark avoidance shuttles in the leaf polysaccharide group(P>0.05).In the Morris water maze experiment,compared with the blank group,the mice in the model group had significantly longer space exploration latency(P<0.01),significantly shorter residence time in the target quadrant(P<0.01),and significantly reduced platform crossing times(P<0.01);Compared with the model group,the spatial exploration latency of the Schisandra root,stem and fructose polysaccharide group was significantly shortened(P<0.05,P<0.01).The stay time in the target quadrant of the mice in the polysaccharide group was significantly prolonged(P<0.01),but there was no significant difference in the stay time in the target quadrant of the root and leaf polysaccharide groups(P>0.05).The number of mice crossing the platform significantly increased in the Schisandra chinensis stem and fructo polysaccharide group(P<0.01).Conclusion The roots,stems and fructose polysaccharides of Schisandra chinensis can improve the learning and memory of aging mice induced by D-galactose.
