摘要
采用组织分离法从贵州安顺发生果斑病的百香果上分离出真菌并纯化,根据柯赫氏法则确定其致病性,明确分离所得菌株B08为致病菌。经形态学观察初步鉴定其为链格孢菌。测定相关基因序列,构建多基因系统发育树,发现B08与梨黑斑链格孢Alternaria gaisen(菌株号:CBS 632.93、LM0519、BMP0243)聚于一支。在室内开展防治药剂的初步筛选,发现氟菌·肟菌酯和苯甲·嘧菌酯对病原菌的抑制效果较好,EC50分别为0.67μg/mL和0.79μg/mL。
The pathogen of passion fruit spot in Anshun of Guizhou was isolated and purified by tissue isolation method.Determined by Koch’s postulates, the isolated strain B08 was identified as the pathogen. It was preliminarily identified as Alternaria by morphological observation. The Alternaria glyceraldehyde-3-phosphate dehydrogenase(gpd), translation elongation factor 1-alpha(TEF1-α), RNA polymerase Ⅱ second largest subunit(RPB2), allergen Alt-a1(Alt-a1), and Calmodulin(Cal) were sequenced, and the phylogenetic tree of multi-gene was constructed. The phylogenetic tree analysis showed that the pathogenic strain B08 and Alternaria gaisen(Strain number: CBS 632.93, LM0519, BMP0243) clustered into one clade. The results of laboratory fungicides screening revealed that the fluopyram·trifloxystrobin and difenoconazole·azoxystrobin had the best inhibitory effects on the pathogen, with EC50values of 0.67 μg/mL and 0.79 μg/mL, respectively.
