摘要
研究旨在补充完善庆阳驴的种质资源特性,建立庆阳驴血液生化指标参考值,开展庆阳驴精液品质检测、冷冻精液制作及血液生化指标检测。试验选取3~4岁庆阳驴种公驴5头、母驴8头,采集精液、血液,开展冷冻精液制作,分别对采精量、精液密度、原精活力、平衡后活力、平衡时间、解冻后活力和23项血清生化指标进行检测分析。结果显示:种公驴平均采精量为66.90 mL,原精活力为0.67,原精密度为1.89亿/mL,平衡时间为2.63 h,平衡后活力0.53,解冻后活力0.37。种公驴采精量与原精液密度呈极显著性正相关(P<0.01),与平衡后活力呈显著正相关(P<0.05)。原精活力、解冻后活力与平衡后活力均呈显著正相关(P<0.05)。所测定的23项血清生化指标中,除公驴血清肌酸激酶(CK)外,其余22项指标的结果与参考值相符。公驴血清白蛋白(ALB)和尿素(UREA)含量极显著高于母驴(P<0.01);葡萄糖(GLU)含量、CK活性及白球比(A/G)显著高于母驴(P<0.05)。母驴血清总胆汁酸(TBA)含量极显著高于公驴(P<0.01),低密度脂蛋白胆固醇(LDL-C)含量显著高于公驴(P<0.05)。研究表明,对庆阳驴精液品质和血清生化指标进行检测可有效指导驴的个体选择与选配,有助于对其开展健康评估,促进种质资源保护和利用。
The purpose of the study was to supplement and improve the characteristics of Qingyang donkey germplasm resources,establish reference values of blood biochemical indicators of Qingyang donkeys,and carry out semen quality testing,frozen semen production and blood biochemical indicators testing of Qingyang donkeys.Five Qingyang male donkeys and eight female donkeys aged from 3~4 years old were selected for the experiment.Semen and blood were collected and frozen semen was prepared.Semen collection,semen density,original semen vitality,post balance vitality,balance time,post thaw vitality and23 serum biochemical indicators were detected and analyzed respectively.The results showed that the average semen yield of male donkeys was 66.90 m L,the original sperm activity was 0.67,the original sperm density was189 million/m L,the balance time was 2.63 h,the balance activity was 0.53,and the thaw activity was 0.37.There was a extreme positive correlation between the amount of semen collected and the original semen density(P<0.01),and there was a significant positive correlation between the amount of semen collected and the vigor after balance(P<0.05).There were significant positive correlations between prospermatogenic activity,post-thaw activity and post-equilibrium activity(P<0.05).Among the 23 serum biochemical indexes measured,except the serum CK of male donkey,the results of the other 22 indexes were consistent with the reference values.The serum ALB and UREA content of male donkeys were extremely higher than those of female donkeys(P<0.01),GLU content,CK activity and A/G were significantly higher than those of female donkeys(P<0.05).The content of TBA in the serum of female donkeys was extremely higher than that of male donkeys(P<0.01),and the content of LDLC was significantly higher than that of male donkeys(P<0.05).The study indicates that the detection of semen quality and serum biochemical indicators of Qingyang donkeys can effectively guide the individual selection and mating of donkeys,help to carry out health assessment,and promote the protection and utilization of germplasm resources.
