摘要
目的探讨β-谷甾醇对幽门螺杆菌感染性胃炎小鼠炎症影响。方法小鼠分成空白组、模型组、低剂量实验组(2 mg·kg^(-1)β-谷甾醇处理)、中剂量实验组(4 mg·kg^(-1)β-谷甾醇处理)、高剂量实验组(6 mg·kg^(-1)β-谷甾醇处理)、对照组(枸橼酸铋钾/替硝唑/克拉霉素处理)。以苏木精-伊红染色评价胃黏膜损伤评分,检测胃黏膜尿素酶情况,以酶联免疫吸附法检测血清中白细胞介素-8(IL-8),以蛋白质印迹法检测诱导型一氧化氮合酶(iNOS)、核因子-κBp65(NF-κBp65)和Toll样受体2(TLR2)蛋白的表达情况。结果空白组、模型组、低剂量实验组、中剂量实验组、高剂量实验组和对照组小鼠的胃黏膜损伤评分分别为0.64±0.50,6.55±0.52,3.36±0.67,2.09±0.30,1.45±0.52和1.55±0.52,血清IL-8水平分别为(26.94±1.63),(2132.17±212.48),(1652.01±162.25),(808.56±80.72),(311.37±20.42)和(757.93±60.52)pg·mL^(-1),iNOS蛋白相对表达水平分别为0.33±0.02,0.84±0.09,0.52±0.06,0.42±0.04,0.52±0.05和0.66±0.06,NF-κBp65蛋白相对表达水平分别为0.33±0.02,0.96±0.08,0.59±0.05,0.47±0.03,0.39±0.04和0.53±0.03,TLR2蛋白相对表达水平分别为0.26±0.04,0.90±0.11,0.72±0.03,0.53±0.06,0.30±0.02和0.52±0.06。模型组的上述指标与空白组比,差异均有统计学意义(均P<0.05);低、中、高剂量实验组和空白组的上述指标与模型组比,差异均有统计学意义(均P<0.05);中、低剂量实验组比,高、中剂量实验组比,差异均有统计学意义(均P<0.05)。结论β-谷甾醇减轻幽门螺杆菌感染性胃炎小鼠胃组织炎症,机制可能与TLR2-NF-κB有关。
Objective To investigate the effect ofβ-sitosterol on inflammation in mice with Helicobacter pylori infectious gastritis.Methods Mice were divided into blank group,model group,low-dose experimental group(2 mg·kg^(-1)β-sitosterol treatment),mediumdose experimental group(4 mg·kg^(-1)β-sitosterol treatment),highdose experimental group(6 mg·kg^(-1)β-sitosterol treatment),control group(potassium bismuth citrate/tinidazole/clarithromycin treatment).Hematoxylin-eosin staining was used to evaluate gastric mucosal injury score,gastric mucosal urease was detected,enzyme-linked immunosorbent assay(ELISA)method was used to detect serum interleukin-8(IL-8),Western blot was used to dectected the expression of inducible nitric oxide synthase(i NOS),nuclear factor-κBp65(NF-κBp65),Toll-like receptors 2(TLR2)protein.Results The gastric mucosal injury scores of the blank group,model group,low-dose experimental group,medium-dose experimental group,high-dose experimental group,and control group were 0.64±0.50,6.55±0.52,3.36±0.67,2.09±0.30,1.45±0.52,1.55±0.52,the IL-8 levels in serum were(26.94±1.63),(2132.17±212.48),(1652.01±162.25),(808.56±80.72),(311.37±20.42),(757.93±60.52)pg·mL^(-1);i NOS protein expression were 0.33±0.02,0.84±0.09,0.52±0.06,0.42±0.04,0.52±0.05,0.66±0.06;the protein expression of NF-κBp65 were 0.33±0.02,0.96±0.08,0.59±0.05,0.47±0.03,0.39±0.04,0.53±0.03;the TLR2protein expression were 0.26±0.04,0.90±0.11,0.72±0.03,0.53±0.06,0.30±0.02,0.52±0.06,the difference of the above indicators between model group and blank group were statistically significant(all P<0.05);the difference between low-dose experimental group,medium-dose experimental group,high-dose experimental group,control group and model group were statistically significant(all P<0.05);the difference between mediumdose experimental group and low-dose experimental group,between high-dose experimental group and mediumdose experimental group were statistically significant(all P<0.05).Conclusionβ-sitosterol reduces gastric inflammation in mice with Helicobacter pylori infectious gastritis,and the mechanism may be related to TLR2-NF-κB.
