不同嘌呤含量食源性低聚肽对高尿酸血症的作用研究 被引量:1

摘要 目的测定不同食物来源食源性低聚肽的嘌呤含量,研究不同嘌呤含量食源性低聚肽对高尿酸血症大鼠模型血尿酸水平、肾功能和肾脏结构的影响。方法通过高效液相色谱测定10种不同取材的食源性低聚肽进行嘌呤含量,大豆肽为高嘌呤,绿豆肽和豌豆肽为中嘌呤,海洋鱼骨肽、海洋鱼蛋白肽、玉米肽、小麦肽、大米肽、白蛋白肽和核桃肽为低嘌呤。选取大豆肽、绿豆肽和海洋鱼蛋白肽为高、中、低嘌呤食源性低聚肽作为大鼠灌胃干预物质。10%果糖构建高尿酸血症大鼠模型;实验分为对照组、模型组、别嘌呤醇组和高、中、低嘌呤含量食源性低聚肽灌胃组,分别在建模7 d、14 d、21 d和28 d取血检测各组大鼠血尿酸、血肌酐和血尿素氮,在28 d实验结束后取肾组织通过HE染色观察肾脏的组织学改变。结果高嘌呤含量的大豆肽在实验4个时间点具有降低血尿酸的作用(P<0.05),建模7 d、14 d、21 d、28 d,模型组血尿酸值分别为(98.7±4.3)μmol/L、(95.6±2.3)μmol/L、(95.2±2.3)μmol/L、(60.15±14.5)μmol/L;高嘌呤大豆肽组血尿酸值分别为(72.4±19.2)μmol/L、(71.0±12.7)μmol/L、(75.6±6.8)μmol/L、(36.81±6.1)μmol/L;且明显改善肾脏损伤、并且降低血肌酐和尿素氮(P<0.05),建模7 d、14 d、21 d、28 d,模型组血肌酐值分别为(26.9±1.1)μmol/L、(27.3±2.0)μmol/L、(28.1±0.9)μmol/L、(39.79±3.5)μmol/L,高嘌呤大豆肽组血肌酐值分别为(24.2±2.4)μmol/L、(23.1±1.5)μmol/L、(23.5±1.2)μmol/L、(29.67±2.3)μmol/L;建模7 d、14 d、21 d、28 d,模型组血尿素氮值分别为(15.9±2.2)mmol/L、(14.4±2.6)mmol/L、(14.9±0.5)mmol/L、(21.60±3.5)mmol/L,高嘌呤大豆肽组血尿素氮值分别为(9.5±4.1)mmol/L、(5.5±2.3)mmol/L、(5.2±2.6)mmol/L、(17.09±1.3)mmol/L。但是中低嘌呤食源性低聚肽仅在建模7 d与大豆肽作用相似,在建模后期无降低尿酸、改善肾损伤的作用;低嘌呤的海洋鱼蛋白肽在建模14 d还有升高血尿酸的作用。结论食源性低聚肽的嘌呤含量与来源食物并不一致。大豆食源性低聚肽能够持续降低模型大鼠的血尿酸、减轻肾脏损伤和肾脏功能异常。 Objective To determine the purine content of foodborne oligopeptides from different food sources and to study the effects of foodborne oligopeptides with different purine contents on blood uric acid level,renal function and renal structure in hyperuricemia rats.Methods The purine content of 10 different kinds of foodborne oligopeptides was determined by high performance liquid chromatography.Soybean peptide was high purine,mung bean peptide and pea peptide was medium purine,and marine fish bone peptide,marine fish protein peptide,corn peptide,wheat peptide,rice peptide,albumin peptide and walnut peptide were low purine.Soybean peptide,mung bean peptide and Marine fish protein peptide were selected as high,medium and low purine foodborne oligopeptides as gavage intervention substances in rats.Hyperuricemia rat model was constructed with 10%fructose.The experiment was divided into control group,model group,allopurinol group and food-borne oligopeptide groups with high,medium and low purine content.Blood samples were collected to detect uric acid,creatinine and urea nitrogen on 7 d,14 d,21 d and 28 d after modeling,and kidney tissues were collected and stained with HE to observe the histological changes of kidneys at 28 d after the experiment.Results Soybean peptide with high purine content had the effect of reducing blood uric acid at four time points in the experiment(P<0.05).On the 7th,14th,21st and 28th day of modeling,the blood uric acid values of the model group were(98.7±4.3)μmol/L,(95.6±2.3)μmol/L,(95.2±2.3)μmol/L and(60.15±14.5)μmol/L;The blood uric acid values of the high purine soybean peptide group were(72.4±19.2)μmol/L,(71.0±12.7)μmol/L,(75.6±6.8)μmol/L and(36.81±6.1)μmol/L;The renal injury was significantly improved,and the blood creatinine and urea nitrogen were decreased(P<0.05).On the 7th,14th,21st and 28th days of modeling,the blood creatinine values in the model group were(26.9±1.1)μmol/L,(27.3±2.0)μmol/L,(28.1±0.9)μmol/L and(39.79±3.5)μmol/L,the blood creatinine value of high purine soybean peptide group was(24.2±2.4)μmol/L,(23.1±1.5)μmol/L,(23.5±1.2)μmol/L and(29.67±2.3)μmol/L;On the 7th,14th,21st and 28th days of modeling,the blood urea nitrogen values in the model group were(15.9±2.2)mmol/L,(14.4±2.6)mmol/L,(14.9±0.5)mmol/L and(21.60±3.5)mmol/L,respectively.The blood urea nitrogen values in the high purine soybean peptide group were(9.5±4.1)mmol/L,(5.5±2.3)mmol/L,(5.2±2.6)mmol/L and(17.09±1.3)mmol/L,respectively.However,the effect of low and medium purine foodborne oligopeptides was similar to that of soybean peptides only after 7 days of modeling,and there was no effect of reducing uric acid and improving kidney damage in the late modeling period;The marine fish protein peptide with low purine also had the effect of increasing blood uric acid after 14 days of modeling.Conclusion The purine content of foodborne oligopeptides is not consistent with the source food.Soybean foodborne oligopeptides can continuously reduce the blood uric acid,alleviate kidney injury and renal dysfunction in model rats.
