摘要
为建立细胞工厂筛选方法,比较市售三个厂家(Corning、Thermo、兰州百灵)细胞工厂培养Vero细胞的生长增殖情况.通过Vero细胞光学显微镜观察、生长曲线、贴壁率、流感病毒增殖对比指标,比较三个厂家细胞工厂培养Vero细胞增殖效果.结果显示,光学显微镜观察三个不同来源的细胞工厂培养的Vero细胞形态变化无明显生长差别,连续传代培养Vero细胞,细胞生长良好.三种不同细胞工厂均能较好促进细胞的生长增殖且细胞生长曲线均呈“S”型;贴壁率分别为(86.20±0.62)%、(94.50±0.70)%、(89.53±0.60)%.甲型H5N1流感病毒感染细胞48 h后,出现明显细胞病变,且血凝效价HA为1∶16,感染性滴度为2.5.三个不同来源的细胞工厂均能支持Vero细胞培养及甲型H5N1流感病毒的增殖,可为细胞工厂筛选提供参考.
Objective To compare the growth and proliferation of Vero cells cultured by three commercial cell factories(Corning,Thermo and Lanzhou Bailing).MethodsThe proliferation effect of Vero cells cultured in three cell factories was compared with observing Vero cells under optical light microscope,growth curve and adhesion rate as well as the comparison index of influenza virus proliferation.Results The results of optical microscopy observation was no significant difference in the growth and morphology of Vero cells cultured in three cell factories.Three different cell factories could promote the cell growth and proliferation.The cell growth curve was"S"type.The adhesion rates were(86.20±0.62)%,(94.50±0.70)%,(89.53±0.60)%,respectively.After 48 hours of infection with influenza A(H5N1)virus,the cells showed obvious cytopathic changes with a hemagglutination titer of 1∶16 and titer of infectivity of 2.5.Conclusion Three cell factories from different sources can support Vero cell culture and proliferation of influenza virus H5N1.
