杧果E2F/DP转录因子家族生物信息学分析

摘要 E2F/DP转录因子是真核生物中细胞周期进程的关键调节因子,与植物激素共同发挥着重要的调控作用。以杧果全基因组为供试参考基因组,用生物信息学软件预测E2F/DP转录因子家族基因成员,并分析其理化性质、保守结构域、保守基序、进化关系,结合实时荧光定量PCR测定杧果细菌性黑斑病菌(Xanthomonas campestris pv.mangiferaeindicae,Xcm)、胶孢炭疽菌(Colletotrichum gloeosporioides,Cg)、茉莉酸甲酯(methyl jasmonate,MeJA)和水杨酸(salicylic acid,SA)处理下基因表达量。结果表明,杧果全基因组中有8个E2F/DP转录因子,命名为MiE2Fs。保守结构域预测发现MiE2F1~MiE2F5均含有E2F_DD,MiE2F7和Mi E2F8同时包含两个E2F_TDP保守结构域;二级结构预测发现E2F/DP家族蛋白质二级结构相似,无规则卷曲和α-螺旋是MiE2F1~MiE2F8蛋白二级结构主要元件,其次是延伸链和β-转角;保守基序预测发现杧果有1个高度保守的基序Motif1;基于杧果和大豆、烟草、毛果杨、拟南芥、阿月浑子、葡萄的E2F/DP蛋白质序列构建系统发育树,发现杧果E2F/DP基因与阿月浑子、葡萄亲缘关系接近。杧果细菌性黑斑病菌(Xcm)和胶孢炭疽菌(Cg)侵染过程中,MiE2F4、MiE2F5、MiE2F6的表达量显著上调。茉莉酸甲酯(MeJA)胁迫处理下,MiE2F7表达量显著上调,其余基因均下调;水杨酸(SA)胁迫处理下,从6 h开始,各个基因的表达量都在逐步上调。以上研究结果可为探究杧果E2F/DP基因参与的植物防御反应机制奠定基础。 E2F/DP transcription factor is a key regulator of cell cycle progression in eukaryotes and plays an important regulatory role together with plant hormones.Using the whole mango genome as a reference genome,we predicted the gene members of E2F/DP transcription by bioinformatics software factor family,and analyzed its physicochemical properties,conserved domain,conserved motif,and evolutionary relationship.The gene expres sion levels of Xanthomonas campestris pv.mangiferaeindicae(Xcm),Colletotrichum gloeosporioides(Cg),methyl jasmonate(MeJA)and salicylic acid(SA)treatment in mango were detected by RT-qPCR.The results showed that there were 8 E2F/DP transcription factors in the whole genome of mango,named MiE2Fs.The conserved domain prediction showed that MiE2F1-MiE2F5 contained E2F_DD,MiE2F7 and MiE2F8 contained two E2F_TDP conserved domains.The prediction of secondary structure showed that E2F/DP family proteins were similar,random coil andα-helix were the main components of secondary structure of MiE2F1-MiE2F8 proteins,followed by extend strand andβ-corner.There was a highly conserved motif in mango.Phylogenetic trees were constructed based on the E2F/DP gene sequences of mango,soybean,tobacco,poplar,Arabidopsis thaliana,pistachio and grape,and it was found that the E2F/DP gene of mango was closely related to pistachio and grape.The expression levels of MiE2F4,MiE2F5 and MiE2F6 were significantly up-regulated during Xcm and CG infection.Under Me JA stress,the expression level of MiE2F7 was significantly up-regulated,while the other genes were down-regulated.Under SA stress,the expression levels of all genes were gradually up-regulated from 6 h.These results may lay a foundation for exploring the mechanism of plant defense response involved in E2F/DP gene in mango.
