摘要
本文主要使用荧光光谱法研究他唑巴坦酸与牛血清白蛋白(BSA)的相互作用机理。通过Stern-Volmer方程、热力学方程以及双对数方程计算两者相互作用的猝灭常数、速率常数、结合位点数、作用力类型等,并研究了金属离子的存在对他唑巴坦酸与BSA的结合的影响。研究结果表明:他唑巴坦酸对BSA的猝灭类型为静态猝灭,它们之间的作用力为疏水作用力。Mn^(2+)、Fe^(3+)、Zn^(2+)的存在对于他唑巴坦酸与BSA的结合有抑制作用,而Co2+的加入增强了他唑巴坦酸与BSA的结合。
The mechanism of action between tazobactam acid and bovine serum albumin(BSA) was studied by fluorescence spectrometry and UV-Vis Spectrophotometry. The quenching constant, rate constant, number of binding bit, and type of action were calculated by Stern-Volmer equation, thermodynamic equation and log-log equation. The effect of metal ions on the binding of tazobactam acid with BSA was also studied. The results showed that the quenching type of tazobactam acid on BSA was static quenching, and the acting force between them was hydrophobic. Mn^(2+), Fe^(3+)and Zn^(2+)inhibited the binding of tazobactam acid to BSA, while Co^(2+)enhanced the binding of tazobactam acid to BSA.
