摘要
研究甘草分别和番红花、侧柏叶提取物组合应用时,对酪氨酸酶二酚酶活性的抑制以及抑制剂间的相互作用。采用酪氨酸酶L-DOPA速率氧化法,评价抑制剂对酪氨酸酶活性的影响和组合物的协同效应,利用DPPH来评价组合物的抗氧化效果。结果显示在等效于酶活性抑制IC50时,甘草与番红花提取物质量比为1∶7时,相互作用指数为0.41;甘草与侧柏叶提取物质量比为3∶10时,相互作用指数为0.52,两个组合物均具有DPPH清除能力。本研究说明甘草分别和番红花、侧柏叶提取物组合应用时,对酪氨酸酶活性的抑制表现为协同效应,协同程度与二者之间的比例有关。
This study aimed to investigate the inhibition effect of combinations of Glycyrrhiza Uralensis with Crocus Sativus and Thuja Orientalis leaf extracts on tyrosinase diphenolase activity, and the interaction index between them. The effect of inhibitors on tyrosinase activity and synergistic effects among them was evaluated by measuring the oxidation rate of L-DOPA. DPPH was used to detect the antioxidant activity of combinations. The results indicated that in 50 %inhibitory rate of tyrosinase activity, the interaction index was 0.41 when Glycyrrhiza Uralensis combined with Crocus Sativus at the ratio 1∶7;the interaction index was 0.52 when Glycyrrhiza Uralensis combined with Thuja Orientalis leaf extract at the ratio 3∶10, both the combinations had DPPH scavenging potential.Inhibition effects of the mixtures on tyrosinase showed synergism when Glycyrrhiza Uralensis combined with Crocus Sativus and Thuja Orientalis leaf extract,respectively, the mixture ratio has a primary influence on the synergistic effects.
