摘要 基于网络药理学探究陈皮治疗心血管疾病的作用机制.依托TCMSP数据库、DTPS靶点预测系统筛选陈皮活性成分及其潜在作用靶点,从GeneCards、DisGeNET、OpenTargets等数据库收集心血管疾病治疗靶点,将2类靶点进行交互处理得到陈皮治疗心血管疾病的靶点;借助Cytoscape 3.7.2软件构建活性成分-靶点网络;应用String平台构建靶蛋白互作网络;利用ClueGO插件富集靶基因GO生物学类型;运用DAVID(Version 6.8)平台分析靶基因KEGG通路,结合Omicshare数据库绘制KEGG通路气泡图.陈皮11个药效成分作用于PPARG、IL1B、ICAM1、VCAM1、PLG、APOB、F2、SOD1等32个心血管疾病治疗靶点;参与活性氧生物合成、脂质代谢正调控、血液凝固、平滑肌细胞增殖调控、活性氮代谢、氧化还原酶活性调控、血压调节等,涉及血脂代谢、心血管调节、血凝调控、炎症、内分泌相关的16条通路.陈皮通过抗炎、抗氧化、抗凝、保护心肌以及调节血脂等药理作用治疗心血管疾病. Based on network pharmacology,the mechanism of Citri Reticulatae Pericarpium in treating cardiovascular diseases was explored.Based on TCMSP database and DTPS target prediction system,the active ingredients and potential targets of Citri Reticulatae Pericarpium were screened,the cardiovascular disease treatment targets were collected from GeneCards,DisGeNET,OpenTargets and other databases,and the two kinds of targets were processed interactively to obtain the target of Citri Reticulatae Pericarpium for cardiovascular disease.The active ingredient target network was constructed with the help of Cytoscape 3.7.2 software.The target protein interaction network was constructed by using String platform.The biological type of target gene GO was enriched by ClueGO plug-in.The target gene KEGG pathway was analyzed by using of DAVID(Version 6.8)platform,and KEGG pathway bubble diagram was drawn combined with Omicshare database.The 11 medicinal components of Citri Reticulatae Pericarpium act on 32 cardiovascular disease treatment targets including PPARG,IL1B,ICAM1,VCAM1,PLG,APOB,F2,SOD1,they participate in the biosynthesis of reactive oxygen species,positive regulation of lipid metabolism,blood coagulation,smooth muscle cells proliferation regulation,reactive nitrogen metabolism,oxidoreductase activity regulation,blood pressure regulation,and it involves 16 pathways related to lipid metabolism,cardiovascular regulation,blood coagulation regulation,inflammation and endocrine.Citri Reticulatae Pericarpium can treat cardiovascular diseases through anti-inflammatory,anti-oxidant,anti-coagulation,myocardial protection,blood lipid regulation and other pharmacological effects.
