摘要
采用MTT法和细胞黏附研究鬼针草提取物对KYSE150增殖和黏附的影响,通过荧光定量PCR技术分析促凋亡基因Bak、Bax、Fas、P53的mRNA表达量的变化,采用蛋白免疫印迹法检测鬼针草提取物对凋亡相关蛋白(BAX)表达量的影响.结果表明,鬼针草提取物能够抑制食道癌细胞的增殖,降低癌细胞黏附作用、破坏细胞间的信息交流,其抑制作用呈现剂量依赖关系;同时,鬼针草提取物可刺激促凋亡基因Bak、Bax、Fas、P53的mRNA表达,促使BAX蛋白表达增加,呈现剂量依赖关系,进而引起细胞凋亡.可见,鬼针草提取物具有抑制食道癌细胞增殖和黏附的作用,并能通过刺激相关促凋亡基因的表达,诱导细胞凋亡.
The MTT assay and cell adhesion were used to study the effect of Bidens Pilosa L.extract on the proliferation and adhesion of KYSE150. The mRNA expression of apoptotic genes Bak,Bax,Fas and P53 were analyzed by real-time PCR. The effect of Bidens Pilosa L. extract on the expression of apoptosis related protein(BAX) was detected by Western blot. The results showed that Bidens Pilosa L. extract could inhibit the proliferation of KYSE150 cells,reduce the adhesion and information exchange of cancer cells in a dose-dependent manner;at the same time,Bidens Pilosa L.extract could stimulate mRNA expression of apoptotic genes Bak,Bax,Fas and P53,increase the expression of BAX protein in a dose-dependent manner,and then cause apoptosis of cells. In conclusion,Bidens Pilosa L. extract could inhibit the proliferation and adhesion of esophageal cancer cells,and induce apoptosis by stimulating the expression of apoptotic genes.
