河南农村汉族人群ABO基因rs507666和rs651007多态性与2型糖尿病的关系

Association of rs507666 and rs651007 of ABO gene polymorphism with type 2 diabetes mellitus in Han population from Henan rural area

摘要 目的:探讨ABO基因rs507666和rs651007多态性及相应的遗传风险评分(GRS)与河南农村汉族人群2型糖尿病(T2DM)患病风险之间的关系。方法:基于“河南农村队列”的基线调查,共纳入648名T2DM病例,按照同性别、年龄±3岁的原则进行1∶1匹配获得648名对照。采用SNPscan®技术对ABO基因rs507666和rs651007进行基因分型。采用条件Logistic回归模型分析ABO基因多态性与T2DM之间的关系。构建3种GRS(简单相加GRS、基于条件Logistic回归的GRS和可释方差GRS)评估其与T2DM患病风险的关联。结果:调整协变量后,相对于基因型GG,rs507666的基因型AA与T2DM的OR(95%CI)为1.794(1.044~3.082);相对于基因型CC,rs651007的基因型TT与T2DM的OR(95%CI)为1.858(1.083~3.185)。rs507666和rs651007的3种GRS与T2DM关联的OR(95%CI)分别为1.109(1.005~1.224)、1.083(1.004~1.168)和1.171(1.007~1.360)。结论:河南农村汉族人群ABO基因rs507666的A等位基因和rs651007的T等位基因及其3种GRS与T2DM患病风险相关。 Aim:To investigate the association of rs507666 and rs651007 of ABO gene polymorphism and the corresponding genetic risk score(GRS) with type 2 diabetes mellitus(T2DM) risk. Methods: A total of 648 T2DM cases from the baseline survey of Henan Rural Cohort were included,and 648 controls were matched according to gender,age ± 3 years. The genotypes of ABO gene polymorphisms were achieved by SNPscan®technology. Conditional Logistic regression model was used to explore association of ABO gene polymorphisms with T2DM risk. Moreover,three GRS methods(SC-GRS,DL-GRS and EV-GRS) were applied. Results: After adjusting for covariates,compared with the genotype GG,the estimated effect[OR(95% CI) ]of genotype AA of the rs507666 locus on T2DM risk was 1. 794(1. 044-3. 082);compared with the genotype CC,the estimated effect[OR(95% CI) ] of genotype TT of the rs651007 locus was 1. 858(1. 083-3. 185). The OR( 95% CI) for the associations of three GRS with T2DM risk were 1. 109(1. 005-1. 224),1. 083(1. 004-1. 168) and 1. 171(1. 007-1. 360),respectively. Conclusion: A allele of rs507666 and T allele of rs651007 of ABO gene as well as three corresponding GRS are associated with the risk of T2DM prevalence in Han population from Henan rural area.
