摘要
探究了对香豆酸(p-coumaric acid,p-CA)对人肺癌细胞细胞A549和H1299增殖、凋亡和迁移的影响及作用机制.结果显示,添加0~10 mM p-CA能显著抑制两种肺癌细胞的生长和迁移,并呈剂量和时间依赖性(P<0.001).p-CA显著抑制了细胞周期发生,并将细胞周期阻滞在G1期,且细胞凋亡发生明显增加(P<0.05).p-CA显著抑制了CDK2、CDK6、Cyclin D1等周期正调控因子表达,并诱导细胞周期负调控因子p21、p27等基因的表达上调;与此同时,p-CA还诱导了促凋亡因子Bax表达上升和抗凋亡因子Bcl-2表达水平的降低且caspase-3和caspase-9的表达上调,而对迁移相关基因表达的检测结果也显示p-CA抑制了β-连环蛋白(β-catenin)的表达并促进钙粘附蛋白E(E-cadherin)表达上升.上述结果表明,p-CA具有抑制肺癌细胞生长、诱导凋亡并抑制其迁移的作用,它主要是通过促进Bax、caspase-3、caspase-9、p21、p27、E-cadherin等蛋白的表达而抑制Bcl-2、Cyclin D1、β-catenin等蛋白的表达发挥其作用.
This study explored the effects of p-coumaric acid(p-CA)on the proliferation,apoptosis and migration of human lung cancer cells A549 and H1299 and its underlying mechanisms.The results showed that adding 0~10 mM P-CA significantly inhibited the growth and migration of the two lung cancer cells in a dose and time dependent manner(P<0.001).P-CA significantly changed the distribution of cell cycle.The percentage of cells in G1 phase significantly increased,and the percentage of apoptotic cells significantly increased also(P<0.05).The expression of cell cycle positive regulators CDK2,CDK6 and Cyclin D1 were significantly inhibited by p-CA,and the expression of cell cycle negative regulators p21&p27 were significantly upgraded.At the same time,p-CA also induced the expression of pro-apoptotic factor Bax and anti-apoptotic factor Bcl-2,and the expression of caspase-3 and caspase-9 were up-regulated.The expression of migration related genes ofβ-catenin and cadherin E were significantly promoted and inhibited by p-CA,respectively.These results suggest that p-CA can inhibit the growth,apoptosis and migration of lung cancer cells,mainly by promoting the expression of Bax,caspase-3,caspase-9,p21,p27&E-cadherin,and by inhibiting the expression of Bcl-2,Cyclin D1 andβ-catenin.
