地仙煎对膝骨关节炎大鼠软骨组织的保护作用

The Protective Effect of Dixianjian on Knee Articular Cartilage Tissue of KOA Rats

摘要 目的:探究药食同源食疗方剂地仙煎对膝骨关节炎(KOA)大鼠软骨组织的保护作用。方法:将50只SD大鼠随机分为正常组、模型组、地仙煎低剂量组(2.2 g/kg)、地仙煎高剂量组(8.8 g/kg)和硫酸氨基葡萄糖组(0.135 g/kg),每组10只。采用前交叉韧带离断法(ACLT)复制大鼠KOA模型,造模后每天灌胃给药1次,连续6周。末次给药12 h后摘取大鼠膝关节,采用苏木精-伊红染色(HE)、番红O固绿染色、Masson染色观察大鼠软骨组织病理变化情况;酶联免疫吸附(ELISA)法检测大鼠血清中蛋白聚糖(PG)、Ⅱ型胶原蛋白(Col-Ⅱ)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)和基质金属蛋白酶-13(MMP-13)表达情况。结果:与模型组相比,地仙煎显著缓解大鼠膝骨关节软骨表面损伤,减轻大鼠软骨表面褶皱、破损和潮线缺损;改善软骨上层和下层失染、染色不均;提高大鼠软骨组织中蛋白聚糖和胶原纤维含量;显著上调大鼠血清中PG和Col-Ⅱ(P<0.01);显著下调大鼠血清中TNF-α(P<0.01)、IL-1β(P<0.05)和MMP-13(P<0.01)。结论:地仙煎能明显改善KOA大鼠软骨组织病理形态,缓解大鼠膝骨关节炎炎症反应,抑制软骨基质降解,对KOA大鼠软骨组织具有明显保护作用。 Objective:To investigate the protective effect of Dixianjian on cartilage tissue of rats with knee osteoarthritis(KOA).Methods:50 SD rats were randomly divided into normal group, model group, Dixianjian group(2.2g/kg),Dixianjian group(8.8g/kg) and glucosamine sulfate group(0.135g/kg),10 in each group.The rat KOA model was replicated using the anterior cruciate ligament dissection method(ACLT),and was administered by gavage once a day for 6 weeks after moulding.The knee joints of the rats were removed 12h after the last administration, and the rats were stained with hematoxylin-eosin staining(HE),hematoxylin-O solid green staining and Masson staining to observe the histopathological changes of the cartilage, ELISA was used to detect collagen type Ⅱ(Col-Ⅱ),proteoglycan(PG),interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α) and matrix metalloproteinase 13(MMP-13) expression in rat serum.Results:Compared with the model group, Dixianjian not only significantly alleviated the damage on the cartilage surface of the knee articular cartilage, as well as the color and shape of the cartilage, but also obviously reduced the folds, damage and the tide line defect of the cartilage and improved the uneven staining.And it increased the content of proteoglycan and collagen fibres in rat cartilage tissues, as well as significantly up-regulated PG and Col-Ⅱ in rat serum(P<0.01),and significantly down-regulated TNF-α(P<0.01),IL-1β(P<0.05) and MMP-13(P<0.01).Conclusion:Dixianjian can significantly improve the histopathological morphology of cartilage in KOA rats, alleviate the inflammatory response of knee osteoarthritis in rats, inhibit the degradation of cartilage matrix, and have significant protective effects on cartilage tissue in KOA rats.
