摘要
为了调查动物园动物禽流感病毒的携带情况,从扬州市动物园采集了106份野禽类新鲜粪便棉拭子样品,进行SPF鸡胚接种、HA/AI试验鉴定、空斑纯化以及HA、NA、PB1、PB2、PA、NP、M、NS共8个片段扩增及测序分析。结果表明,分离到1株H3N2亚型禽流感病毒,分离率为0.94%,命名为A/Swan/Yangzhou/YZS1084/2019(H3N2)。HA基因裂解位点只有1个碱性氨基酸,符合低致病性禽流感的特征;携带242Q和244G的HA受体结合位点具有优先结合唾液酸α-2,3受体的能力,具有典型禽源特征。在扬州首次分离到的该病毒的8个基因片段均属于欧亚分支,与猪源和人源的毒株差异较大;8个基因片段来源均不相同,具有明显的遗传多样性。
To investigate the prevalence of avian influenza virus in zoo animals,106 fresh fecal swabs were collected from wildfowls of Yangzhou zoo in Yangzhou.The samples were identified by SPF chicken embryo inoculation,HA/HI test,virus plaque purification test,and eight-fragment gene amplification and sequencing analysis.The results showed that one H3N2 subtype of avian influenza virus strain was isolated,and the separation rate was 0.94%,it was named as A/Swan/Yangzhou/YZS1084/2019(H3N2).There was only one basic amino acid at the cleavage site of HA gene,which was consistent with the characteristics of low pathogenic avian influenza virus.HA receptor binding sites with 242Q and 244G preferentially bound toα-2,3 sialic acid receptor,which showed typical avian characteristics.All the eight gene fragments of the virus isolated for the first time in Yangzhou belonged to the Eurasian branch,which were quite different from those from pig or human.The sources of eight gene fragments were all different,and the virus strain had obvious genetic diversity.
