摘要
目的:探究藤椒总生物碱的最佳提取工艺条件,并研究藤椒总生物碱对小鼠肝纤维化的保护作用,以期为藤椒作用于肝脏保护提供理论依据。方法:采用醇提法提取藤椒总生物碱,通过单因素和正交试验优化提取参数,并通过动物实验对藤椒总生物碱的抗肝纤维化效果进行评价。结果:总生物碱得率最高的提取参数为:乙醇浓度75%、提取时间1h、提取温度70℃,在此条件下藤椒总生物碱提取率为5.15%。动物实验结果显示,与模型组相比,藤椒总生物碱干预后,小鼠的肝脏系数明显降低,肝组织病理程度减轻,且小鼠血浆的谷草转氨酶Aspartate aminotransferase,AST)、谷丙转氨酶(Alanine aminotransferase,ALT)、碱性磷酸酶(Alkaline phosphatase,ALP)、总胆红素(Total bilirubin,T.BiLi)四项指标均有明显下降。结论:采用醇提法提取藤椒总生物碱对小鼠表现出良好的抗纤维化作用。
Objective:To explore the optimal extraction process conditions of total alkaloids from Tengjiao and to study the protective effect of Tengjiao total alkaloids on liver fibrosis in mice,in order to provide a theoretical basis for the role of Tengjiao in liver protection.Method:The total alkaloids of Tengjiao were extracted by alcohol.The extraction parameters were optimized by single factor and orthogonal test.The anti-liver fibrosis effect of the Tengjiao total alkaloids was evaluated by animal experiment.Result:The results showed that the optimal extraction conditions were as follows:ethanol of 75%,extraction at 70°C for 1h.Under these conditions,the extraction yield of total alkaloids was 5.15%.Animal experimental results showed that compared with the model group,after the total alkaloid intervention,the liver coefficient of mice was significantly decreased,the degree of pathological injury of liver tissue was alleviated and the four indexes of AST,ALT,ALP and T.BiLi in plasma were significantly decreased.Conclusion:Extraction of total alkaloids from Tengjiao by alcohol extraction show a good anti-liver fibrosis ability.
