环状RNA0005654与特异性蛋白1在甲状腺癌患者中表达水平及临床意义的研究 被引量:1

摘要 目的 探究环状RNA0005654(Circ0005654)、特异性蛋白1(SP1)在甲状腺癌患者中的表达情况及临床意义。方法 选择2017年1月—2019年7月在皖西卫生职业学院附属医院及安徽医科大学附属六安医院行根治性手术的甲状腺癌患者76例作为研究对象,术中取甲状腺癌组织及配对癌旁组织,收集患者临床病理资料。甲状腺癌组织与癌旁组织中SP1蛋白表达情况采用免疫组织化学法(IHC)及Western Blot(WB)检测;Circ0005654表达水平利用实时荧光定量聚合酶链反应(qRT-PCR)法进行检测。通过比较癌组织及癌旁组织中Circ0005654、SP1表达的差异性,分析其与甲状腺癌患者临床病理特征之间的关系。统计患者术后3年生存率,分析Circ0005654、SP1水平对患者预后的影响。结果 WB检测显示癌旁组织中SP1表达水平低于癌组织,差异有统计学意义(t=2.795,P=0.019);IHC结果显示,SP1在癌旁组织中的表达阳性率低于癌组织(χ~2=159.61,P<0.001);qRT-PCR检测显示Circ0005654在甲状腺癌组织中的表达水平明显高于癌旁组织[(2.13±0.59)vs(0.67±0.56)],两组比较,差异具有显著统计学意义(t=15.608,P<0.0001)。Circ0005654、SP1表达水平与TC患者肿瘤直径、淋巴结转移、TNM分期和包膜侵犯相关(P均<0.05)。根据甲状腺癌组织中Circ0005654表达水平的不同,高表达组3年生存率为77.5%,低于低表达组的94.4%,差异有统计学意义(χ~2=4.395,P=0.036);SP1高表达组和低表达组患者3年生存率分别为78.6%和100%,两组比较,差异有统计学意义(χ~2=4.973,P=0.049)。结论 Circ0005654与SP1在甲状腺癌组织中均存在高表达,与患者肿瘤直径、淋巴结转移、TNM分期和包膜侵犯相关,Circ0005654及SP1水平越高,生存率越低,可能具有促进甲状腺癌进展作用。 Objective To explore the expression levels of circular RNA0005654(Circ0005654) and specific protein 1(SP1) in thyroid cancer tissues and their clinical significance.Methods A total of 76 patients suffered from thyroid cancer,those were admitted to affiliated hospital of West Anhui Health Vocational College or Lu’an hospital affiliated to Anhui Medical University during January 2017 and July 2019 and received radical operation,were enrolled as study subjects.The thyroid cancer tissue and corresponding adjacent tissues were taken during the radical surgery,and the clinical and pathological data were collected.The expression of SP1 in thyroid cancer tissues and adjacent tissues were determined by immunohistochemical method(IHC) and Western Blot(WB).The real-time quantitative polymerase chain reaction(qRT-PCR) was used to analyze the expression of Circ0005654 in thyroid cancer tissues and adjacent tissues.According to compare the variability of Circ0005654 and SP1 in cancerous and adjacent tissues,the relationship between Circ0005654,SP1 expression levels and the clinicopathological characteristics of thyroid cancer patients was analyzed.The survival rate within 3 years was counted.The effect of Circ0005654 and SP1 levels on the prognosis of the patient were analyzed.Results According the results of WB,the expression levels of SP1 in adjacent cancer tissues were lower than that in thyroid tissues,the difference was statistically significant(t=2.795,P=0.019).The results of IHC staining showed that the positive expression rate of SP1 in adjacent tissues was significantly lower than that in thyroid cancer tissues (^(χ)2 = 159.61,P<0.001).The data of qRT-PCR showed the level of Circ0005654 in thyroid cancer tissues washigher than that in adjacent tissues(2.13±0.59) vs(0.67±0.56),t= 15.608,P<0.0001.The expression levels ofCirc0005654 and SP1 were correlated with the tumor diameter,lymph node metastasis,TNM stage,and capsuleinvasion(allP<0.05).According to the Circ0005654 expression level in the thyroid cancer tissues,the 3-yearsurvival rate in the high expression group was 77.5%,it was lower than 94.4% in the low expression group,thedifference was statistically significant (^(χ)2 = 4.395,P = 0.036).The 3-year survival rate of patients in SP1 highexpression group and low expression group were 78.6% and 100%,respectively,the difference was statisticallysignificant (^(χ)2 = 4.973,P = 0.029).Conclusions Both Circ0005654 and SP1 were highly expressed in thyroidcancer tissues which are associated with tumor diameter,lymph node metastasis,TNM stage and capsule invasion.The higher level of Circ0005654 and SP1,the lower the survival rate,which may promote the progression of thyroidcancer.
