摘要
目的 探讨喉癌组织内微小RNA(miR)-340-5p(miR-340-5p)、原癌基因(c-Myc)与转录因子-4(TCF-4)的表达及其临床意义。方法 选取64例喉癌患者为研究对象。于术中采集癌组织及癌旁正常组织,应用实时荧光定量逆转录聚合酶链反应(RT-PCR)检测癌组织及癌旁正常组织miR-340-5p、c-Myc与TCF-4的表达水平,另外统计分析三项指标表达水平与喉癌患者临床病理特征的关系。结果 癌组织中miR-340-5p相对表达量(2.15±0.27)低于癌旁正常组织(6.76±0.89),c-Myc、TCF-4相对表达量[(4.95±0.63)、(4.36±0.48)]高于癌旁正常组织[(0.94±0.16)、(0.90±0.17)],差异有统计学意义(P<0.05);miR-340-5p、c-Myc、TCF-4与喉癌患者年龄、体重指数(BMI)、肿瘤分化程度、肿瘤位置、是否伴有淋巴结转移无关(P>0.05);miR-340-5p、c-Myc、TCF-4与喉癌患者的肿瘤分期有关(P<0.05)。结论miR-340-5p、c-Myc、TCF-4在喉癌患者癌组织内呈异常表达,其中miR-340-5p呈低表达,c-Myc、TCF-4呈高表达,三项指标与喉癌分期有关,参与喉癌的发生与发展,临床需予以高度重视。
Objective To investigate the expression and clinical significance of microRNA(miR)-340-5p(miR-340-5p),proto-oncogene(c-Myc) and transcription factor-4(TCF-4) in laryngeal carcinoma.Methods 64 patients with laryngeal cancer were selected as the study subjects.The expression levels of miR-340-5p, c-Myc and TCF-4 in cancer tissues and normal tissues adjacent to cancer were detected by real-time fluorescence quantitative reverse transcriptase polymerase chain reaction(RT-PCR).In addition, the relationship between the expression levels of 3 indicators and the clinicopathological characteristics of laryngeal cancer patients was statistically analyzed.Results The relative expression of miR-340-5p in cancer tissue(2.15±0.27) was lower than that in normal tissue adjacent to cancer(6.76±0.89),and the relative expression of c-Myc and TCF-4 [(4.95±0.63),(4.36±0.48)] was higher than that in normal tissue adjacent to cancer [(0.94±0.16),(0.90±0.17)],the difference was statistically significant(P<0.05);MiR-340-5p, c-Myc, TCF-4 were not associated with age, body mass index(BMI),tumor differentiation, tumor location, and lymph node metastasis in laryngeal cancer patients(P>0.05);MiR-340-5p, c-Myc and TCF-4 were related to the tumor stage of laryngeal cancer patients(P<0.05).Conclusion miR-340-5p, c-Myc and TCF-4 are abnormally expressed in laryngeal cancer tissues, of which miR-340-5p is low and c-Myc and TCF-4 are high.The 3 indicators are related to the stage of laryngeal cancer, and are involved in the occurrence and development of laryngeal cancer, which should be highly valued in clinical practice.
