低共熔溶剂温浸提取葡萄皮籽渣中原花青素的工艺优化研究 被引量:3

摘要 目的采用低共熔溶剂温浸提取葡萄皮籽渣中原花青素,优化主要影响因素,建立绿色高效的提取工艺。方法联合单因素考察与中心组合实验设计,以葡萄皮籽渣中原花青素提取含量为评价指标,逐步优选提取溶剂、提取方法以及低共熔溶剂含水量、料液比、提取温度、提取时间等工艺参数。结果提取工艺是以含水量16.47%的氯化胆碱-乳酸(1∶2)为提取溶剂,料液比为1∶10.63,在50℃、磁力搅拌提取40 min条件下原花青素提取含量达143.53 mg·g^(-1),是传统的乙醇回流提取工艺的2.3倍;实验结果与响应面模型预测值接近。结论相较于传统乙醇回流浸提法,低共熔溶剂温浸法提取葡萄皮籽渣中原花青素的效率明显提高。该方法具有简便、高效特点,可为低共熔溶剂提取天然活性产物提供参考。 Objective This study aimed to screen key factors influencing the warm infusion efficacy of proanth-ocyanidin from grape pomace by deep eutectic solvents(DES)and to optimize its process parameters,establish a new green and efficient strategy.Methods Using proanthocyanidin content in grape pomace as the evaluation index,single-factor experiment and central composite design were combined to optimize extraction solvent,extraction method and process parameters including water content of deep eutectic solvent,the ratio of solvent and material,extraction temperature and extraction time.Results Warm infusion method assisted by magnetic force agitation was selected at 50℃for 40 min.The optimized DES was choline chloride and lactic acid at a molar ratio of 1∶2 and with its water content of 16.47%.And the ratio of solvent and material was set at 10.63∶1.Proanthocyanidin content of the optimized DES infusion process reached 143.53 mg·g^(-1),which was 2.3 times of that of ethanol reflux extraction process.The results were close to the predicted values of the response surface methodology.Conclusion The result suggests that compared with traditional ethanol reflux extraction method,the warm infusion efficacy of proanthocyanidin from grape pomace by deep eutectic solvents infusion method is a rapid,simple and efficient approach for extraction of proanthocyanidin from grape pomace.The study provides a reference for the efficient preparation of natural products with green and designable DES solvents.
