摘要
目的:研究五皮饮对淋巴水肿小鼠的影响,并初步探讨其作用机制。方法:选择48只C57BL/6J雄性小鼠,依据随机数字表随机分成空白组,模型对照组,五皮饮高、中、低剂量组和呋塞米组,连续灌胃14 d,观察并记录小鼠尾部的水肿变化情况。灌胃结束后,处死小鼠并取出小鼠远端尾部组织进行检测。苏木精-伊红(hematoxylin-eosin staining,HE)染色法染色观察小鼠尾部横截面的病理学变化;免疫组织化学染色法检测小鼠尾部横截面中同源盒基因转录因子1(prospero homeobox protein 1,Prox-1)的表达水平;免疫组织荧光染色法检测小鼠尾部横截面中血管内皮生长因子受体3(vascular endothelial growth factor receptor-3,VEGFR-3)和淋巴内皮透明质酸受体-1(lymphatic vessel hyaluronan receptor-1,LYVE-1)的表达水平;Western blot法检测小鼠尾部组织中Prox-1、VEGFR-3和LYVE-1的蛋白表达水平。结果:与空白组比较,模型对照组小鼠尾部直径显著增加,皮肤厚度明显增加,水肿区范围增多,毛细血管扩张明显;尾组织横截面切片Prox-1、LYVE-1和VEGFR-3的表达明显降低。与模型对照组比较,五皮饮高、中、低剂量组和呋塞米组小鼠尾部直径减少,水肿区范围变小,Prox-1、VEGFR-3和LYVE-1的表达降低,尤其是五皮饮高剂量组表现最为明显(P<0.05)。结论:五皮饮可以上调Prox-1、VEGFR-3和LYVE-1的表达,通过促进淋巴管新生,从而缓解淋巴水肿。
Objective:To study the effect of Wupi Yin on lymphedema in mice and to explore its mechanism.Methods:48 male C57BL/6J mice were randomly divided into the blank group,the model group,the Wupi Yin groups of high-dose,medium-dose and low-dose,as well as the Furosemide group.After 14 days of intervention,the edema changes around tail were observed and recorded.At the end of gavage,mice were sacrificed and the distal tail tissue of mice was removed for detection.HE staining was used to observe the pathological changes in the tail section of mice.The expression of prospero-related homeobox 1(Prox-1)in the tail section of mice was detected by immunohistochemistry.The expressions of vascular endothelial growth factor receptor-3(VEGFR-3)and lymphatic vessel endothelial hyaluronan receptor-1(LYVE-1)in the tail section of mice were detected by immunofluorescence staining.The protein expressions of Prox-1,VEGFR-3 and LYVE-1 in the tail tissue of mice were detected by Western blot.Results:Compared with those in the blank group,the tail diameter,skin thickness,edema area and telangiectasia were significantly increased in the model group;the expressions of Prox-1,LYVE-1 and VEGFR-3 were significantly decreased in the model group(P<0.05).Compared with those in the model,the tail diameter and edema area were reduced,and the expressions of Prox-1,VEGFR-3 and LYVE-1were decreased in the Furosemide group and the Wupi Yin groups of high-dose,medium-dose and low-dose,especially in the high-dose group(P<0.05).Conclusion:Wupi Yin can up-regulate the expressions of Prox-1,VEGFR-3 and LYVE-1,it can relieve lymphedema by promoting lymphangiogenesis.
