蒲金口服液对HIBD幼鼠脑组织PSD-95、NMDAR-2B表达的影响

摘要 目的探讨蒲金口服液对缺氧缺血性脑损伤(Hypoxic-ischemic brain damage,HIBD)幼鼠的神经保护机制。方法单侧颈总动脉结扎联合缺氧环境制备HIBD幼鼠模型。动物分为中药组、西药组、模型组、假手术组、空白组。中药组蒲金口服液0.1 mL/(kg·d)灌胃;西药组单唾液酸四己糖神经节苷脂钠注射液3 mL/(kg·d)腹腔注射;余组生理盐水0.1 mL/(kg·d)灌胃,均每日1次,连续14 d。Morris水迷宫测试学习与记忆行为能力,尼氏染色观察脑组织神经元,透射电镜观察突触超微结构,Western Blot检测脑组织PSD-95、NMDAR-2B表达。结果蒲金口服液可改善HIBD幼鼠神经损伤,上调脑组织突触后密度蛋白95(PSD-95)、谷氨酸受体2B抗体(NMDAR-2B)表达(P<0.05)。结论蒲金口服液改善HIBD幼鼠神经行为学表现,可能与其调控PSD-95、NMDAR-2B表达,维护突触稳态有关。 Objective To explore the neuroprotective mechanism of Pujin oral liquid in neonate rats with HIBD.Methods The model of neonate rats with HIBD were prepared by unilateral common carotid artery ligation combined with hypoxic environment.According to the model preparation measures,the rats were divided into traditional Chinese medicine(TCM)group,western medicine group and model group,sham group and blank group.The TCM group was administered Pujin oral liquid at 0.1 ml/kg once a day.The western medicine group gave 3 ml/kg ganglioside sodium injection in intraperitoneal injection once a day,rest groups were gavaged with 0.1 ml/kg of normal saline once a daily.All groups were intervened for 14 consecutive days.Learning and memory behavioral ability were performed using the Morris water maze.Neurons of brain tissue were observed by Nissl staining.Synaptic ultrastructure was visualized by transmission electron microscopy.The expression of PSD-95 and NMDAR-2B was determined by Western Blot.Results Pujin oral liquid ameliorated nerve injury in neonate rats with HIBD,up-regulated PSD-95 and NMDAR-2B in brain tissues(P<0.05).Conclusion Pujin oral solution can improve the neurobehavioral performance of neonatal rats with HIBD,this may be related to its regulation of PSD-95 and NMDAR-2B expression to maintain synaptic homeostasis.
