摘要
目的:通过检测斑马鱼对尼可刹米的作用,揭示斑马鱼作为一种新型模式生物用于医学实验教学中的可行性。方法:设定不同浓度的尼可刹米,投放斑马鱼,观察其游泳状态、惊厥反应及死亡情况,并与小鼠惊厥反应对比。结果:尼可刹米溶于水中可以导致斑马鱼游泳行为的改变,表现为不同程度的兴奋、游泳速度变快、摆尾幅度增大、惊厥、侧翻飘浮和死亡,并且动物反应与药物的作用浓度及时间呈正相关;另外,致惊厥量的尼可刹米诱发小鼠惊厥反应,表现为小鼠上蹿下跳、转圈、惊厥和死亡等现象。结论:以斑马鱼为模式生物检测尼可刹米诱发惊厥反应的检测方法安全可靠、操作简单易行,实验结果准确、重复性高,是将来在医学实验教学中用于替代小鼠检测药物毒性反应和对其他医学实验的开展具有潜在应用价值的一种理想实验动物模型。
Objective:The possibility of using zebrafish as a new model animal in medical experiment teaching was verified by detecting the reaction of zebrafish to nikethamide which was dissolved in water.Methods:After being exposed to different concentrations of nikethamide,the swimming state,convulsive reaction and death of zebrafish were observed and compared with those of mice injected with convulsive amount of nikethamide.Results:Nikethamide dissolved in water can cause the occurrence of convulsive reaction in zebrafish.Zebrafish showed excitement and behavioral changes such as increased swimming speed,wagging tail violently,convulsion,rollover and death.Interestingly,the drug response was positively correlated with the drug concentration and action time.In addition,the convulsive amount of nikethamide resulted in convulsive reaction,which was manifested by jumping up and down,circling,convulsion,relaxation and death in mice.Conclusion:Zebrafish is a safe,reliable,simple and easy-to-operate animal model for detecting convulsion induced by nikethamide.It is an ideal experimental animal model to substitute mice for detecting convulsion reaction and other pharmacological experiments in medical experimental teaching in the future.
