电感耦合等离子体质谱(ICP-MS)法测定黄精中15种稀土元素及其指纹图谱绘制 被引量:4

摘要 为了准确测定黄精中稀土元素的含量,采用微波消解法处理黄精样品后,通过在线引入内标溶液,用电感耦合等离子体质谱(ICP-MS)法同时测定黄精中15种稀土元素(La、Ce、Pr、Nd、Pm、Sm、Eu、Gd、Tb、Dy、Ho、Er、Tm、Yb、Lu)的含量,15种稀土元素的线性关系、精密度良好,平均加标回收率在97.0%~103%,相对标准偏差不大于1.7%。采用OriginPro 2021绘制稀土元素指纹图谱和进行主成分分析,以稀土元素含量的平均值构建黄精稀土元素标准指纹图谱,作为鉴定黄精中药材的参考依据;采用SPSS 26.0进行聚类分析。聚类分析结果表明,当平方欧式距离为10时,浙江产地样品(S5~S8)聚为一类,其余的聚为一类,与主成分分析结果一致。黄精药材中15种稀土元素的指纹图谱具有相似的分布形态,具有较强的特征性和一致性,相似度均大于0.950。黄精中Ce、La、Nd含量相对较高,其中Ce含量最高为53.0~2005μg/kg。方法操作简单,准确可靠,能满足实验分析要求,可为黄精的质量控制和药理研究提供参考,建立的指纹图谱可用于黄精的鉴别。 To accurately determine the content of rare earth elements in Polygonati Rhizoma,the samples were digested by microwave with the introduction of the online internal standard solution, 15 rare earth elements(La, Ce, Pr, Nd, Pm, Sm, Eu, Gd, Tb, Dy, Ho, Er, Tm, Yb, Lu) in Polygonati Rhizoma were determined by inductively coupled plasma mass spectrometry(ICP-MS),the 15 rare earth elements showed the good linearity and precision, and the average recoveries were between 97.0%—103%,while the relative standard deviations were less than or equal to 1.7%.The rare earth elemental fingerprint and principal component analysis were performed using OriginPro 2021 software, the rare earth standard elemental fingerprint was established by using the average concentrations of each rare earth element, which could be used as reference for the identification of Polygonati Rhizoma herbal medicine.Clustering analysis was performed using SPSS 26.0 software.The results of clustering analysis showed that the samples(S5-S8) of Zhejiang province origin was grouped into one class, and the rest were grouped into one class, which was consistent with principal component analysis.The fingerprint of 15 rare earth elements in Polygonati Rhizoma showed a similar element spectrum, and its spectrum had strong characteristics and consistency while the similarity was greater than 0.950.The content of Ce, La, and Nd in Polygonati Rhizoma was relatively higher than the other elements, and the content of Ce was 53.0—2 005 μg/kg, ranking first.This method is simple to operate, accurate and reliable, which can meet the requirements of experimental analysis and provide the reference for quality control and pharmacological research of Polygonati Rhizoma;The established fingerprint can be used for the identification of Polygonati Rhizoma.
