不同采样法对洁净室表面微生物检测效果的比较研究

摘要 目的:通过不同采样方法对模拟洁净室不同表面材料微生物检测结果的研究,寻求最佳的洁净室表面微生物采样法。方法:比较3种采样法对4种载体片材料上5种标准菌(金黄色葡萄球菌、铜绿假单胞菌和枯草芽孢杆菌3种细菌,白色念珠菌和黑曲霉2种真菌)回收率的影响,评估载体材料和采样方法对洁净室表面微生物检测效果的影响。结果:采用表面擦拭-涂布采样法对模拟洁净室环境采样,在聚四氟乙烯载体材料上菌种的平均回收率最高;采用接触皿采样法,玻璃载体材料的平均回收率最高;采用表面擦拭-水浸泡采样法时,聚四氟乙烯的平均回收率最高。结论:洁净室表面微生物采样方法应根据不同的室内表面材料选择,对于玻璃等相对光滑的载体表面应采用接触皿采样法,而对于聚四氟乙烯等粗糙的载体表面应采用表面擦拭-水浸泡法。 Objective:By investigating the influence of different carriers and different sampling methods on the sampling results of clean room surface microorganism,to find the best method of clean room surface microorganism sampling.Methods:Through the analysis and comparison of the average recovery rate of Staphylococcus aureus,Pseudomonas aeruginosa and Bacillus subtilis represented by bacteria and Candida albicans and Aspergillus Niger represented by fungi under three sampling methods,to evaluatethe influence of the carrier and method on the surface microorganism of clean room.Results:The average recovery rate of bacteria on the PTEE carrier is the best when the surface wiping-coating sampling method is used to sample the simulated clean room environment.The average recovery rate of glass carrier is the best when the contact plate sampling method is adopted.The average recovery of PTE carrier is the best when the method of surface swabbing and water soaking is used.Conclusion:The method of microbial sampling on clean room surface should be selected according to different indoor surface materials.Contact plate sampling method should be used for carrier surface like glass,while surface wiping and water soaking method should be used for carrier surface like PTEE.
