摘要
以马桑菌(Lentinus edodes)菌株LeM-f为试材,采用外源添加GABA并覆盖赛璐玢膜的平板置于室温和亚温逆境下培养,观测不同浓度γ-氨基丁酸(GABA)对菌丝生长速度、气生菌丝鲜质量、菌丝生长抑制率、显微形态及抗氧化酶活性变化,以期为探索应用GABA对缓解马桑菌菌丝对亚温胁迫的逆境耐受能力、抗性提升效果提供参考依据。结果表明:外源添加适宜浓度GABA能缓解菌丝遭受的亚高、低温胁迫,促进菌丝生长,提高菌丝密度,且增加了前端菌丝的分支率,抗氧化酶(CAT、SOD)活性随外源添加GABA而提升。0.6 mmol·L^(-1)GABA对亚高温胁迫的缓解效果最佳,生长抑制率低至8.63%,接近室温正常生长水平,气生菌丝产量比亚高温抑制时提升212.47%,分支率提高34.31%,所有效果优于亚低温胁迫。综上所述,外源GABA可效缓解亚高、低温造成的伤害,但亚高温条件下作用最佳,生产中的应用前景很广。
Lentinus edodes strain LeM-f was used as test material,plates covered with celluloid membrane were cultured at room temperature and subthermal stress to observe the changes of different concentrations of GABA on the growth rate of mycelia,fresh weight of aerogenic mycelia,inhibition rate of mycelia growth,microscopic morphology and antioxidant enzyme activity,in order to provide reference for exploring the effects of GABA on alleviating the stress tolerance and resistance enhancement effect of Lentinus edodes mycelium to subtemperature stress.The results showed that exogenous addition of appropriate concentration of GABA could alleviate the sub-high and low temperature stress suffered by hyphae,promote the growth of mycelium,increase hyphae density,and increase the branching rate of front-end hyphae,and the activity of antioxidant enzymes(CAT,SOD)increased with the addition of exogenous GABA.GABA of 0.6 mmol·L^(-1)had the best alleviation effect on sub-high stress with a growth inhibition rate as low as 8.63%,which was close to the normal growth level at room temperature,and the yield of aerial mycelium was increased by 212.47%and the branching rate was increased by 34.31%compared with sub-high temperature inhibition,all effects were better than sub-low temperature stress.In summary,exogenous GABA could effectively alleviate the damage caused by sub-high and low temperature,but it had the best effect under sub-high temperature conditions,and the application prospect in production was very wide.
