苦豆碱在艾滋病患者外周血PBMC中调控Wnt/β-catenin信号通路影响T细胞表达机制分析 被引量:1

摘要 目的观察苦豆碱干预艾滋病患者外周血单个核细胞(peripheral blood mononuclear cell,PBMC)后,对Wnt/β-catenin信号通路相关蛋白水平及T细胞表达的影响,从而探讨苦豆碱治疗艾滋病的机制。方法选取广州医科大学附属市八医院感染科收治的20名艾滋病患者作为试验者,随机分为实验组及对照组。分离试验者外周血PBMC,分别加入苦豆碱溶液及0.9%氯化钠溶液干预培养,干预前后用流式细胞仪检测CD3^(+),CD4^(+),CD8^(+),CD45^(+)和Foxp3^(+)表达;Western blot检测苦豆碱对糖原合成酶激酶-3β(glycogen synthase kinase-3β,GSK-3β),磷酸化糖原合成酶激酶-3β(phospho-GSK-3β,p-GSK-3β),β-连环蛋白(β-catenin),磷酸化-β-连环蛋白(phospho-β-catenin,p-β-catenin),蛋白磷酸酶2A(protein phosphatase 2A,PP2A)和叉头转录因子(forkhead transcription factor p 3,Foxp3)蛋白水平的影响。结果经过苦豆碱的干预,实验组CD3^(+),CD4^(+)和Foxp3^(+),CD4^(+)/CD8^(+)比值上升,CD8^(+)表达下降,较对照组差异有统计学意义(P<0.05)。苦豆碱干预后,实验组p-GSK-3β,β-catenin,PP2A和Foxp3蛋白水平上升,GSK-3β和p-β-catenin水平下降(P<0.05,P<0.05)。结论苦豆碱能有效上调Wnt/β-catenin信号通路,促进艾滋病患者外周血PBMC的CD4^(+)T和Foxp3^(+)细胞表达,有助于临床治疗,PP2A可能是治疗靶标。 Objective To observe the effect of aloperine on T cells expressionand Wnt/β-catenin signaling pathway regulationin AIDS patients'peripheral blood mononuclear cell(PBMC),so as to find out the therapeutic mechanism of aloperine in AIDS.Methods 20 AIDS patients were divided into experimental group and control group randomly,and their PBMCs were isolated.Cells were cultured with aloperine and 0.9%Nacl respectively.The levels of CD3^(+),CD4^(+),CD8^(+),CD45^(+)and Foxp3^(+)were detected by flow cytometry before and after culture.The protein of GSK-3β,p-GSK-3β,β-catenin,p-β-catenin,PP2A and Foxp3were detected by Western blot.Results Compared with the control group,aloperine increased the ratio of CD3^(+),CD4^(+),Foxp3^(+)and CD4^(+)/CD8^(+)in the experimental group and decreased the ratio of CD8^(+)(P<0.05).Moreover,aloperine up-regulated the protein level of p-GSK-3β,β-catenin,PP2A,Foxp3^(+)and down-regulated the proteinlevel of GSK-3β,p-β-catenin(P<0.05,P<0.05).Conclusion Aloperine can effectively up-regulate Wnt/β-catenin signaling pathway by increasing the level of CD4^(+)T cells and Foxp3^(+)cells in PBMC of AIDS patients.PP2A may be an important therapeutic target.
