摘要
为解决咖啡胚性愈伤悬浮培养增殖效率较低、悬浮增殖培养获得的胚性细胞在后续体胚转化率低的问题,以中粒种咖啡叶片诱导的胚性愈伤组织为材料,建立了咖啡胚性愈伤悬浮培养体系。在此基础上,分别探讨了起始胚性愈伤培养时间、继代起始浓度以及培养基中不同激素水平等条件对悬浮培养及后续体胚转化率(胚性保持)的影响。结果表明,起始胚性愈伤培养时间为3周,继代起始浓度为0.03mL胚性细胞/1mL培养液,细胞生长素2,4-D与细胞分裂素KT比例为1∶1时,净增殖率最高且胚性愈伤组织后续的体胚诱导率相对较高。探讨了咖啡胚性愈伤悬浮培养的适宜条件,对构建咖啡胚性愈伤悬浮培养系,提高组织培养种苗繁育效率具有一定意义。
In order to solve the problem of low proliferation efficiency of coffee embryogenic callus suspension culture and low transformation rate of embryogenic cells obtained from suspension culture in subsequent somatic embryos,the embryogenic callus induced from the leaves of Coffea canephora was used as the material in this experiment to establish the coffee embryogenic callus suspension culture system.On this basis,the effects of initial embryogenic callus culture time,subculture initial concentration and different hormone levels in the medium on suspension culture and subsequent somatic embryo transformation rate(embryogenic preservation)were discussed.The results showed that when the initial embryogenic callus culture time was 3 weeks,the subculture initial concentration was 0.03mL embryogenic cells/1mL culture medium,and the ratio of cytokinin 2,4-D to cytokinin KT was 1∶1,the suspension proliferation efficiency was the highest and the subsequent somatic embryo transformation rate of embryogenic callus was relatively high.This study discussed the suitable conditions of coffee embryogenic callus suspension culture,which was of certain significance to construct coffee embryogenic callus suspension culture system and improve the efficiency of tissue culture seedling breeding.
