摘要
目的:建立一种基于G4/hemin过氧化物模拟酶快速降解罗丹明B的方法。方法:重复的富含G碱基的结构单元的DNA片段能形成堆积的G-四链体结构(G4),分子内平行G4-DNA与氯化血红素室温孵育成强活性的过氧化物模拟酶。吸取罗丹明B溶液于具塞比色皿中,依次加入过氧化物模拟酶、过氧化氢溶液,以手摇方式进行降解反应。结果:加入过氧化氢30 min后,溶液的颜色由桃红色变为酒红色,通过罗丹明B标准曲线计算得至少90%以上的罗丹明B染料已降解。结论:该方法具有成本较低,方法简便,稳定性和重复性良好等优点,可推广至环境中罗丹明B的快速降解。
Objective:To establish a rapid degradation of Rhodamine B method based on G4/hemin mimetic peroxidase.Methods:Repeated G-base-rich DNA fragments can form a stacked G-quadruplex structure(G4).Highly active peroxidases were formed by incubating intramolecular parallel G4-DNA with hemin at room temperature.Rhodamine B,mimetic peroxidase and hydrogen peroxide solutions were added into a cuvette with stopper successively and degradated with shaking hands.Results:The color of the solution changed from pink to wine red,after adding hydrogen peroxide 30 mins.At least 90%of Rhodamine B dye was degraded by Rhodamine B standard curve.Conclusion:The established method is simple,cheap,stable and repeative.It can be used to degradate Rhodamine B in environment.
