多房棘球蚴单克隆抗体的免疫特性分析

摘要 用免疫学方法对2株抗多房棘球蚴单克隆细胞株进行免疫特性分析。研究制备抗多房棘球蚴单克隆抗体1G11、1C1,亲和层析纯化并检测纯度,采用ELISA、Western blot和免疫组化法分别检测所获抗体的抗体效价、特异性以及抗体在免疫组织化学定位。结果显示,经亲和层析纯化获得的单克隆抗体1G11、1C1在分子质量约55 ku及24 ku可见清晰的重、轻链条带;ELISA与Western blot结果显示,单克隆抗体1G11、1C1与多房棘球蚴能特异性结合,与细粒棘球蚴抗原、细颈囊尾蚴抗原、多头蚴抗原不反应且无交叉反应;经免疫组化鉴定2株抗多房棘球蚴单克隆抗体均能与多房棘球蚴特异性结合。结果表明,抗多房棘球蚴单克隆抗体具有良好的免疫特性,为后期包虫病诊断试剂的研发提供了物质基础。 Analysis of two anti-Echinococcus multilocularis monoclonal cell lines was conducted by immunological method.The prepared anti-Echinococcus multilocularis monoclonal cell lines 1Gll and 1C1 were purified by affinity chromatography and followed by the purification determination.The antibody titer,antibody specificity and immunohistochemical localization of the antibodies were detected by ELISA,Western blot and immunohistochemical method respectively.The results revealed that light chain band and heavy chain band of the monoclonal antibodies 1Gl1 and 1C1,with purification through affinity chromatography,were clearly discernible at the relative molecular weights of about 55 ku and 24 ku;The results of ELISA and Western blot demonstrated that monoclonal antibodies 1Gll and 1C1 can realize the specific binding between itself and Echinococcus multilocularis,without reaction and cross-reactivity with Echinococcus granulosus antigen,Coenurus cerebralis antigen and Cysticercus tenuicollis antigen;The determination results gained through immunohistochemistry method showed that two anti-Echinococcus multilocularis monoclonal antibodies could specifically bind to Echinococcus multilocularis.To summarize,hybridoma cell lines 1Gll and 1Cl of anti-Echinococcus multilocularis monoclonal antibodies can provide stable secretion of antibodies and favorable immune characteristics,enabling a material basis for further research and development of hydatidosis diagnostic reagent.
