响应面法优化菟丝子类药材总黄酮提取工艺及含量测定 被引量:8

Optimization of extraction technology and content determination of total flavonoids of Cuscutae spp.Semen by response surface methodology

摘要 为优化菟丝子总黄酮提取工艺条件,本试验对69批不同地区来源的菟丝子类药材进行含量测定并比较差异。运用NaNO2-Al(NO3)3-NaOH比色法测定总黄酮含量;利用单因素试验考察乙醇浓度、料液比、提取温度对菟丝子总黄酮含量的影响;采用Box-Behnken Design试验(以下简称BBD试验)设计原理进行三因素三水平试验设计及响应面分析来优化菟丝子总黄酮提取工艺。结果表明:菟丝子总黄酮最佳提取工艺为乙醇浓度65%,料液比1:41(g/mL),提取温度92℃;不同地区来源的小菟丝子总黄酮含量为0.92%~2.18%,大菟丝子总黄酮含量为2.57%~3.34%。Box-Behnken Design响应面优化菟丝子总黄酮的提取工艺可靠,具有实用价值;大菟丝子与小菟丝子能否同用值得进一步研究;建议《中国药典》增加菟丝子药材以芦丁(C27H30016)计,含总黄酮不得低于0.73%。 To optimize the extraction conditions of total flavonoids from Cuscutae Semen,and to determine the content of 69 batches of Cuscuta spp.Semen from different regions and compare the content differences.NaNO2-Al(NO3)3-NaOH colorimetric method was used to determine the content of total flavonoids.The effects of ethanol concentration,solid-liquid ratio and extraction temperature on the extraction yield of total flavonoids from Cuscutae Semen were investigated by single factor experiment.Box-Behnken Design(BBD)was used to optimize the extraction process of total flavonoids from Cuscutae Semen.The optimum extraction conditions were as follows:ethanol concentration 65%,solid-liquid ratio 1:41(g/mL),extraction temperature 92℃.The content of total flavonoids in Cuscutae Semen from different regions ranged from 0.92%to 2.18%,and the content of total flavonoids in Cuscutae Japonicae Semen ranged from 2.57%to 3.34%.The optimization of Box-Behnken Design response surface for extracting total flavonoids from Cuscutae Semen was reliable and has practical value.Whether Cuscutae Japonicae Semen and Cuscutae Semen could be used together was worth further study.It was suggested that the total flavonoids content of Cuscutae Semen in terms of rutin(C27H30O16)should not be less than 0.73%in Chinese Pharmacopoeia.
