皖南牛mtDNA D-loop区遗传多样性和系统进化分析 被引量:1

摘要 采用DNA提取及PCR扩增技术,对30头皖南母牛的mtDNA D-loop区的全序列进行扩增与分析。利用Genbank数据库,下载国内外部分黄牛品种mtDNA D-loop区的全序列,再使用Clustalx、Dnasp5.0等生物软件,对其全序列进行比对和对单倍型多样度、核苷酸多样度等展开分析。皖南牛mtDNA D-loop区序列长度均在962~1052 bp的区间内,共发现68个变异位点,其中单一信息位点共有18个,简约信息位点共有50个。构成13种单倍型,其单倍型多样性(Hd)的均值为0.6483,核苷酸多样性(Pi)的均值为0.0131,其序列之间核苷酸的差异平均数(k)为11.589,A、C、G、T碱基含量平均占比分别为33.26%,25.23%,13.39%,28.12%。通过遗传距离和N-J系统进化树得知,皖南牛和威宁黄牛、湘西牛、大别山牛和吉安黄牛亲缘关系较近。皖南牛的遗传多样性和群体变异程度较低。研究结果可为皖南牛遗传资源的保护开发提供科学依据和理论参考。 DNA extraction and PCR amplification were used to amplify and analyze the D-loop sequence of mtDNA from 30 head of Wannan cattle.The sequence alignment and analysis of haplotype diversity and nucleotide diversity were carried out by using Clustalx>Dnasp5.0 and other biological software,combined with the full sequence of mtDNA D-loop area downloaded from the Genbank database.The results showed that the sequence length of the mtDNA D-loop region of Wannan cattle was found to be in the range of 962 to 1052 bp with 68 variable sites,including 18 single information cites and 50 parsimony informative sites.There were 13 haplotypes,and the haplotype diversity(Hd)was found.The mean value was 0.6483,the mean value of nucleotide diversity(Pi)was 0.0131,and the mean value of nucleotide difference(k)between sequences was 11.589,and the contents of A,C,G and T bases were 33.26%,25.23%,13.39%and 28.12%respectively.The data indicated that the genetic diversity of southern Anhui cattle was abundant,and the variation of population was similar to that of other Chinese cattle population.Based on genetic distance and N-J phylogeny,Wannan cattle are genetically close to Weining,Xiangxi,Dabieshan and Ji'an cattle.Wannan cattle have low genetic diversity and population variation.The results could provide scientific basis and theoretical reference for protection and development of Warman cattle resources.
