基于50K SNP芯片的夏洛来羊遗传结构及选择信号分析 被引量:3

摘要 为了分析中国夏洛来羊繁育群的遗传结构及其在引入我国后因环境和人为因素所发生的改变,试验利用绵羊50K单核苷酸多态性(single nucleotide polymorphism, SNP)芯片对75只中国夏洛来羊进行了SNP检测,分析中国夏洛来羊繁育群的亲缘关系及家系构成;同时将50K芯片数据与24只法国夏洛来羊芯片数据进行整合,分析中国和法国夏洛来羊群体遗传多样性及选择信号。结果表明:75只中国夏洛来羊的状态同源(identity by state, IBS)遗传距离在0.149 8~0.337 1之间,大部分个体之间的遗传距离较远,划分为5个家系;与法国夏洛来羊相比,中国夏洛来羊群体遗传多样性相对较低;从75只中国夏洛来羊基因组中筛选到8个候选SNPs位点(chr5:96209980、chr3:102592698、chr16:27817793、chr19:41609459、chr10:25335663、chr10:25565841、chr22:30954815、chr1:116966008),这些位点分别在乳蛋白率、产奶量、乳脂率等产奶性状和最大日增重、体重、胴体重、体内脂肪含量等生产性状及鸡粪便虫卵计数、蛇形毛圆线虫等抗病性状相关的数量性状基因座(quantitative trait locus, QTL)上,筛选出LOC101113975、C3H2orf81、LOC114115134、LHFPL6、NHLRC3、PROSER1、PTPRG 7个候选基因。说明中国夏洛来羊群体结构基本合理,绝大多数个体遗传距离较远,且中国夏洛来羊和法国夏洛来羊之间已产生了一定程度的遗传分化。 This study aimed to analyze the genetic structure of Chinese Charolais sheep breeding population and its changes due to environmental and artificial factors after introduction into China.SNP detection was performed on 75 Chinese Charolais sheep by sheep 50K single nucleotide polymorphism(SNP) chip to analyze the genetic relationship and family composition of breeding population. At the same time, the 50 K chip data were integrated with the chip data of 24 French Charolais sheep to analyze the genetic diversity and selection signals of Chinese and French Charolais sheep populations. The results showed that the identity by state(IBS) genetic distance of 75 Chinese Charolais sheep was between 0.149 8 and 0.337 1, and the genetic distance between most individuals was far, which was divided into 5 families. Compared with the local Charolais sheep in France, the genetic diversity of Chinese Charolais population was relatively low. Eight candidate SNP loci(chr5:96209980, chr3:102592698, chr16:27817793, chr19:41609459, chr10:25335663, chr10:25565841, chr22:30954815, chr1:116966008) were screened from the genome of 75 Chinese Charolais sheep. These loci were located on the quantitative trait locus(QTL) related to milk production traits such as milk protein rate, milk yield, milk fat rate, and QTL related to production traits such as maximum daily gain, body weight, carcass weight, body fat content, and QTL related to disease resistance traits such as fecal worm egg count in chickens and trichostrongylus colubriformis. Seven candidate genes such as LOC101113975, C3H2orf81, LOC114115134, LHFPL6, NHLRC3, PROSER1 and PTPRG were screened. The results indicated that the population structure of Chinese Charolais sheep was basically reasonable, the genetic distance of most individuals was far, and a certain degree of genetic differentiation had been generated between Chinese Charolais and French Charolais.
