摘要
骨关节炎(osteoarthritis,OA)是世界上最常见的关节疾病,人群发病率约10%[1]。其特征性结构改变包括滑膜炎症、关节软骨丢失、软骨下骨改变、半月板损伤以及肌腱和韧带退化[2]。OA是老年人关节疼痛、功能丧失和致残的最主要原因,其中膝关节受影响最大[3]。目前OA的发病机制尚未明确,但现有的研究表明,炎症、焦亡与凋亡在OA的疾病进展中发挥着重要作用[4-5]。
Osteoarthritis( OA) is the leading cause of pain, loss of function and disability in the elderly,affecting more than 780 million people around the world. Nucleotide-binding oligomerization domain-like receptor protein 3( NLRP3) has been proved to be related to the pathological process of many diseases. In particular, recent studies have shown that the activation of NLRP3 inflammasome is related to the pathogenesis and progression of OA.In this review, we will introduce the characteristics of molecular structure and signal activation mechanism of NLRP3inflammasome, elaborate the role of NLRP3 in the pathological process of OA, and discuss potential therapeutic strategies targeting NLRP3 for OA treatment.
