摘要
朝鲜崖柏(Thuja koraiensis)体内含有挥发性化合物,具有药用价值,研究挥发性化合物的相关基因对了解其合成十分重要.使用气相色谱-质谱(GC-MS)技术分析朝鲜崖柏体内挥发性化合物,根据转录组数据的代谢途径确定与挥发性物质相关的基因;用定量PCR(qRT-PCR)研究挥发性物质相关基因的表达模式.共鉴定出84种挥发性化合物.结果表明:Kaur-16-烯,(8β,13β)-(53.472%)、双环[9.3.1]十五碳-3,7-二烯-12-醇、4,8,12,15,15-五甲基-[1R-(1R*,3E,7E,11R*,12R*)]-(5.698%)、十六烷(5.197%)和1,1′-联苯,2,2′,5,5′-四甲基-(4.918%)是朝鲜崖柏的主要化合物,并发现10种化合物具有同分异构体;代谢途径分析表明,与Kaur-16-烯和角鲨烯合成相关的14个基因被注释到两个代谢途径上,这14个基因涉及ent-copalyl二磷酸酯合成酶、ent-kaurene合成酶和角鲨烯合成酶;与5月的基因表达量相比,所有的基因在6月、7月和8月均上调表达,而在1月、2月、3月、4月、10月、11月和12月下调表达.研究结果揭示了朝鲜崖柏挥发性化合物的生物合成途径,有助于朝鲜崖柏的有效利用.
Korean arborvitae(Thuja koraiensis)is a valued medicinal species for its volatile compounds.It is important to explore volatile compounds and volatiles-associated genes to understand volatile compounds biosy-nthesis in Korean arborvitae.We used gas chromatography-mass spectrometry(GC-MS)technology to analyze the volatile compounds according to the pathway analysis of transcriptome data and volatiles-associated genes.Quantitative PCR(qRT-PCR)was used to investigate the expression patterns of volatiles-associated genes.84 volatile compounds were identified.Kaur-16-ene,(8β,13β)-(53.472%),bicycle[9.3.1]pentadeca-3,7-dien-12-ol,4,8,12,15,15-pentamethyl-[1R-(1R*,3E,7E,11R*,12R*)]-(5.698%),hexadecane(5.197%)and 1,1′-biphenyl,2,2′,5,5′-tetramethyl-(4.918%)were main compounds,and ten compounds had isomers in the extracts.Pathway analysis identified that kaur-16-ene and squalene,and fourteen genes were assigned two pathways.The fourteen genes were involved in ent-copalyl diphosphate synthase,ent-kaurene synthase and squalene synthase.Compared with the original gene in May,all the genes were up-regulated in June,July and August,and were down-regulated in January,February,March,April,October,November and December.These results showed volatile compounds biosynthesis pathway and helped effective utilization on Korean arborvitae.
