巴东地区鲊广椒细菌多样性分析及乳酸菌分离鉴定

摘要 该研究使用高通量测序技术对巴东地区鲊广椒中细菌多样性进行解析,同时使用传统纯培养方法对其蕴含的乳酸菌菌株进行分离鉴定。结果表明,14个鲊广椒样品中细菌隶属于23个门下的304个属,2个平均相对含量>1.0%的门为厚壁菌门(Firmicutes)(96.45%)和变形菌门(Proteobacteria)(2.12%),3个平均相对含量>1.0%的属为乳酸杆菌属(Lactobacillus)(92.22%)、魏斯氏菌属(Weissella)(2.19%)和葡萄球菌属(Staphylococcus)(1.48%)。从14个鲊广椒样品中共分离出58株乳酸菌,其中53株被鉴定为植物乳杆菌(L. plantarum)、消化乳杆菌(L. alimentarius)、戊糖乳杆菌(L. pentosus)、仙人掌乳杆菌(L. senmaizukei)、短乳杆菌(L. brevis)和副干酪乳杆菌(L. paracasei),5株被鉴定为屎肠球菌(Enterococcus faecium)。L. plantarum和L. alimentarius占分离株总数的51.72%和25.86%。由此可见,巴东地区鲊广椒细菌多样性较高,且其乳酸杆菌以L. plantarum和L. alimentarius为主。 Bacterial diversity of Zha-chili in Badong region was analyzed with high throughput sequencing technology,and the lactic acid bacteria strains were isolated and identified with traditional pure culture method.The results showed that the bacteria from 14 Zha-chili samples belonged to 304 genera under 23 phyla,and the two phyla with average relative content of greater than 1.0%were Firmicutes(96.45%)and Proteobacteria(2.12%).Three genera with an average relative content of greater than 1.0%were Lactobacillus(92.22%),Weissella(2.19%)and Staphylococcus(1.48%).A total of 58 strains of lactic acid bacteria were isolated from 14 Zha-chili samples,and 53 strains were identified as Lactobacillus plantarum,L.alimentarius,L.pentosus,L.senmaizukei,L.brevis and L.paracasei,5 strains were identified as Enterococcus faecium,L.plantarum and L.alimentarius accounted for 51.72%and 25.86%of total isolates.In conclusion,bacteria diversity of Zha-chili in Badong region was higher,and Lactobacillus were dominated by L.plantarum and L.alimentarius.
