谷氨酸棒杆菌硫酯酶基因NCgl1600的异源表达及酶学性质研究

摘要 硫酯酶在脂肪酸合成中具有解除脂酰CoA的抑制及提高脂肪酸产量的作用。因谷氨酸棒杆菌具有脂肪酸合成潜力,故文章以具有脂酰CoA硫酯酶活性的基因NCgl1600作为研究对象,通过异源表达基因NCgl1600获得其表达产物硫酯酶TesB并研究其酶学性质。通过测定底物特异性发现其对C8脂肪酰基CoA底物活性最高,最适温度为30℃,最适pH值为8.0,金属离子Fe^(2+)、Cu^(2+)、Mn^(2+)、Zn^(2+)、Mg^(2+)和K^(+)对其酶活产生抑制作用,米氏常数Km为56.81 mmol/L。 Thioesterase can relieve the inhibition of fatty acyl CoA and improve the yield of fatty acid in fatty acid synthesis.Because Corynebacterium glutamicum has the potential of fatty acid synthesis,this study took the gene NCgl1600 with fatty acyl CoA thioesterase activity as the research object,obtained its expression product thioesterase TesB through heterologous expression gene NCgl1600,and its enzymatic properties were studied.By determining the specificity of the substrate,it was found that it had the highest activity against C8 fatty acyl CoA substrate.The optimum temperature was 30 C,the optimum pH value was 8.0,the metal ions Fe^(2+)、Cu^(2+)、Mn^(2+)、Zn^(2+)、Mg^(2+)and K^(+) inhibited the enzyme activity,andtheMichaelisconstantKmwas56.81mmol/L.
