摘要
9-羟基-雄烯二酮(9a-OH-AD)是一种重要的甾体药物中间体,能够合成多种皮质类甾体激素药物。本文以公司保藏的分枝杆菌Mycobacterium sp.DR01为出发菌株,通过紫外和亚硝基胍复合诱变,筛选得到一株突变菌株Mycobacterium sp.DR11,然后以植物甾醇为底物,以9a-OH-AD产量为衡量指标,通过对培养基、转化条件等参数进行优化,以及15L发酵罐小试和100L发酵罐中试放大试验,实现了20t发酵罐在植物甾醇投料量6%情况下转化96h,底物转化率达95%以上的无油化工业生产水平。
9a-OH-AD is an important steroid drug intermediate,which can synthesize a variety of corticosteroid drugs.In this study,Mycobacterium sp.DR01 preserved by the company was taken as the original strain,through UV and nitrosoguanidine compound mutagenesis,a mutant strain Mycobacterium sp.DR11 was screened,then plant sterols are used as substrates,taking the output of 9a-OH-AD as the measurement index,through the optimization of culture medium,transformation conditions and other parameters,as well as the small-scale test of 15L fermentor and the pilot scale test of 100L fermentor,the 20t fermentation tank is realized under the condition that the feeding amount of phytosterol is 6%,conversion 96h,the production level of oil-free chemical industry with substrate conversion rate of more than 95%.
