基于微流控芯片技术的瘀血痹胶囊治疗类风湿关节炎处方分析

Prescription analysis of Yuxuebi capsules for rheumatoid arthritis based on microfluidic chip technology

摘要 目的基于微流控芯片技术,进行瘀血痹胶囊治疗类风湿关节炎处方分析,为瘀血痹胶囊的物质基础研究提供实验依据。方法以瘀血痹胶囊11味中药为研究对象,基于微流控芯片技术设计并制作治疗类风湿关节炎的药效筛选芯片,按均匀设计方法所得22个不同配伍组进行给药干预,以细胞凋亡率为评价指标,建立回归方程得到瘀血痹胶囊药效最佳的药物组合及其最优配比,并与原方进行药效差异性分析。结果与空白对照组比较,瘀血痹胶囊各配伍组均能诱导RA-FLSs细胞凋亡(P<0.01)。根据逐步回归分析结果,乳香、没药、姜黄、丹参药物组合诱导RA-FLSs细胞凋亡效果最佳,最佳配比为乳香(X_(1))∶没药(X _(2))∶姜黄(X_(10))∶丹参(X_(11))=2.628∶2.946∶4.208∶6.852。将筛选出的复方与瘀血痹胶囊进行药效差异性对比,在相同剂量时,4味中药按最优配比得到的复方(简称最优配伍方),与按原方比例仅含4味中药配伍的复方(简称筛选方)诱导细胞凋亡作用相当,且筛选方能够达到瘀血痹胶囊作用的77%左右。结论本研究基于微流控芯片技术,得到乳香、没药、姜黄、丹参的药物组合是瘀血痹胶囊处方中发挥治疗类风湿关节炎作用的主要组成。 Objective To analyze the prescription of Yuxuebi capsules for rheumatoid arthritis to provide material basis for research by microfluidic chip technology.Methods Totally 11 traditional Chinese medicines in Yuxuebi capsules were selected as the research object.A pharmacodynamic screening chip for rheumatoid arthritis was designed and produced based on microfluidic chip technology,and 22 matching groups were intervened with uniform design method.The apoptosis rate was taken as the evaluation index,the regression equation was established to determine the best efficacy of Yuxuebi capsules for drug combinations and their optimal ratios,and the efficacy difference was analyzed with the original formula.Results Compared with the blank control group,all compatible groups of Yuxuebi capsules induced the apoptosis of RA-FLSs cells(P<0.01).According to stepwise regression analysis,the combination with the best effect on inducing the apoptosis of RA-FLSs cells consisted of olibanum,myrrha,curcumae longae and salvia miltiorrhiza,and the optimal ratio was olibanum(X_(1))∶myrrha (X_(2))∶curcumae longae (X_(10))∶salvia miltiorrhiza (X_(11)) = 2.628∶2.946∶4.208∶6.852. The efficacy difference between the screened compound and Yuxuebi capsules was compared: at the same dose, the compound obtained according to the optimal ratio of the 4 traditional Chinese medicines (referred to as the optimal compatible formula) showed similar apoptosis induction with the compound containing only 4 traditional Chinese medicines in the original ratio (referred to as the screened formula);and the screened formula reached about 77% of the effect of Yuxuebi capsules. Conclusion Drug combination of olibanum, myrrh, curcumae longae and salvia miltiorrhiza based on microfluidic chip technology, proves to be the main components in Yuxuebi capsules prescription for rheumatoid arthritis.
