摘要
目的探讨红景天苷对白血病人急性T淋巴细胞白血病CEM细胞凋亡过程的影响。方法用3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑溴盐法[3-(4,5-Dimethylthiazol-2-yl)-2,5diphenyltetrazoliumbromide,MTT]法检测红景天苷对CEM细胞生长能力的作用;倒置显微镜和Hoechst33258染色分别观察红景天苷作用CEM细胞后形态的变化;流式细胞术测定红景天苷对CEM细胞凋亡的影响;Westernblot法测定有关凋亡蛋白的表达水平。结果红景天苷可以剂量和时间依赖方式显著抑制CEM细胞的生长,其作用24、48、72h后的(IC_(50))分别为40.020、11.160、9.396 mmol/L;与对照组比较,红景天苷组细胞数量显著下降,结构也被破坏,并出现了明显的细胞凋亡形态;流式结果显示随着红景天苷浓度的升高,凋亡率逐渐升高;红景天苷可以上调Bax、细胞色素C(cytochromec,Cyt-C)、半胱氨酸蛋白酶3(cysteinyl aspartate specific proteinase3,Caspase-3)和Caspase-9的表达,而下调B淋巴细胞瘤-2蛋白(B-cell lym-phoma-2,Bcl-2)的表达。结论红景天苷可通过线粒体途径促使CEM细胞凋亡。
Objective To discuss the influence and mechanism of salidroside on the apoptotic process of leukemia CEM cells.Methods The effect of salidroside on CEM cell growth capacity was detected with MTT method.Inverted microscope and Hoechst 33258 staining were used to observe the changes of CEM cells after salidroside intervention.Flow cytometry was used to detect the influence of apoptosis of CEM cells after salidroside intervention.Western Blot method assayed the expression of apoptosis protein.Results Salidroside can significantly inhibit the growth of CEM cells in a dose and time-dependent manner.The IC_(50) values after 24,48 and 72 h of its action were 40.020,11.160 and 9.396 mmol/L,respectively.Compared with those in the control group,the number of cells in salidroside group decreased significantly,the structure was also destroyed and there was obvious apoptosis.Flow results showed that with the elevation of the salidroside concentration,the apoptotic rate was ascended obviously.Salidroside could promote the expressions of Bax,Cyt-C,Caspase-3 and Caspase-9,but restrain the expression of Bcl-2.Conclusion Salidroside can cause CEM cell apoptosis by mitochondrial pathway.
