摘要 目的:探讨肝细胞癌组织中白细胞介素4诱导蛋白1(IL4I1)的表达及其临床价值。方法:从TCGA公共数据库中下载肝细胞癌的相关样本资料,使用R软件和Perl软件进行数据分析整理,采用Wilcoxon秩和检验分析基因IL4I1在正常组织和肝细胞癌组织中的差异表达以及与临床病理特征的相关性,经单因素和多因素Cox回归分析肝癌患者预后的影响因素。以基因集富集分析(GSEA)方法分析基因IL4I1的调控网络,探究潜在机制。计数资料采用中位数(四分位数间距)表示。结果:基因IL4I1在肝细胞癌组织中的表达量显著高于正常组织﹝0.69(0.31,1.75)比0.22(0.15,0.51),P<0.05﹞。IL4I1表达量与肿瘤的组织学分级﹝G_(1)、G_(2)、G_(3)、G_(4)分别为0.31(0.19,0.76)、0.75(0.32,1.78)、0.93(0.42,2.14)、0.86(0.46,2.07),P<0.01﹞以及TNM分期中的T分期﹝T_(1)、T_(2)、T_(3)、T_(4)分别为0.66(0.30,1.52)、0.92(0.36,2.79)、0.64(0.26,1.92)、1.57(0.84,2.91),P<0.05﹞呈正相关。GSEA提示,IL4I1参与了自然杀伤细胞介导的细胞毒性作用、细胞因子信号通路、抗原处理和提呈途径、T细胞受体信号通路、JAK-STAT信号通路和Notch信号通路。结论:IL4I1在肝细胞癌组织中呈高表达,与患者的不良预后密切相关,其可能通过JAK-STAT信号通路和Notch信号通路促进肝细胞癌的发生和发展。 Objective:To investigate the expression and clinical value of interleukin-4-induced protein 1(IL4I1)in hepatocellular carcinoma.Method:The relevant sample data of hepatocellular carcinoma were downloaded from the TCGA public database.R software and Perl software were used for data analysis and collation.Wilcoxon rank sum test was used to analyze the differential expression of IL4I1 gene in normal tissues and hepatocellular carcinoma tissues and its correlation with clinicopathologic features.Univariate and multivariate Cox regression were used to analyze the prognostic factors of liver cancer patients.The regulatory network of gene IL4I1 was analyzed by gene set enrichment analysis(GSEA)to explore the potential mechanism.Count data were expressed as median(interquartile).Results:The expression level of IL4I1 in hepatocellular carcinoma tissues was significantly higher than that in normal tissues(0.69(0.31,1.75)vs.0.22(0.15,0.51),P<0.05).The expression of IL4I1 and histological grade of tumor(G_(1),G_(2),G_(3),G_(4) were 0.31(0.19,0.76),0.75(0.32,1.78),0.93(0.42,2.14),0.86(0.46,2.07),respectively.P<0.01)and T stages of TNM stages(T_(1),T_(2),T_(3) and T_(4) were 0.66(0.30,1.52),0.92(0.36,2.79),0.64(0.26,1.92),1.57(0.84,2.91),P<0.05),respectively.GSEA suggests that IL4I1 is involved in natural killer cell-mediated cytotoxic effects,cytokine signaling pathways,antigen processing and presentation pathways,T cell receptor signaling pathways,JAK-STAT signaling pathways,and Notch signaling pathways.Conclusion:IL4I1 is highly expressed in hepatocellular carcinoma,which is closely related to poor prognosis of patients,and may promote the occurrence and development of hepatocellular carcinoma through JAK-STAT signaling pathway and Notch signaling pathway.
