广西盾翅藤花芽分化过程及内源激素含量变化研究 被引量:4

摘要 为探究广西盾翅藤花芽分化进程中形态结构变化与内源激素间的关系,以不同时期的花芽为试材,通过石蜡切片和徒手解剖法观察分化过程中的花芽形态变化,并使用HPLC-MS/MS液质联用法对花芽内源激素含量进行测定。结果显示:(1)广西盾翅藤花芽分化可划分为花原基分化期、萼片原基分化期、花瓣原基分化期、花器官原基分化期、花器官形成期以及花器官成熟期共6个时期。(2)广西盾翅藤花芽分化过程中,GA_(3)、BR整体呈下降趋势,GA_(4)、Me-JA含量呈波浪状变化趋势,IAA、ABA含量先下降后升高,ZR、IPA含量呈“降-升-降”的趋势;ZR/GA_(3)、ZR/IAA、GA_(3)/IAA比值先升高再下降,ABA/GA_(3)、(IAA+ZR)/GA_(3)、(GA_(3)+IAA+ZR)/ABA比值先下降后上升。研究结果表明:低水平的GA_(3)、BR、ABA、IAA、Me-JA和高比值的ZR/GA_(3)、ZR/IAA、GA_(3)/IAA有利于花芽分化启动;低水平GA_(4)和高水平的ZR、ABA/GA_(3)、(IAA+ZR)/GA_(3)、(GA_(3)+IAA+ZR)/ABA有利于花器官的形成与发育。广西盾翅藤花芽分化与内源激素关系密切,受多种激素共同调控,研究结果可为花期调控提供科学依据。 In order to investigate the relationship between morphological and structural changes and endogenous hormones in flower bud differentiation of A.concava,the morphological changes during differentiation were observed by paraffin sectioning and freehand dissection,and the endogenous hormone contents of flower buds were determined by HPLC-MS/MS.The results showed that the flower bud differentiation of A.concava can be divided into six periods:floral primordial differentiation,sepal primordial differentiation,petal primordial differentiation,floral organ primordial differentiation,floral organ formation and floral organ maturation.In the process of flower bud differentiation,GA_(3) and BR showed an overall decreasing trend;GA_(4) and Me-JA contents showed a wave-like trend;IAA and ABA contents first decreased and then increased;ZR and IPA contents showed a decreasing-increasing-decreasing trend;ZR/GA_(3),ZR/IAA and GA_(3)/IAA ratios first increased and then decreased,and ABA/GA_(3),(IAA+ZR)/GA_(3),(GA_(3)+IAA+ZR)/ABA ratios decreased and then increased.The results showed that low levels of GA_(3),BR,ABA,IAA,Me-JA and high ratios of ZR/GA_(3),ZR/IAA,GA_(3)/IAA were favorable for the initiation of floral bud differentiation;low levels of GA_(4) and high levels of ZR,ABA/GA_(3),(IAA+ZR)/GA_(3),(GA_(3)+IAA+ZR)/ABA were favorable for the formation and development of floral organs.The floral bud differentiation of A.concava is closely related to endogenous hormones and is regulated by multiple hormones together,which can provide a scientific basis for floral regulation.
