燃煤氟对雌性大鼠下丘脑、垂体、卵巢脏器系数及组织TGF-β1表达的影响

摘要 目的 观察燃煤氟中毒模型雌鼠下丘脑、垂体、卵巢脏器系数变化及组织转化生长因子β1(TGF-β1)表达情况,探讨氟对雌性大鼠性腺轴功能影响的可能机制。方法 120只雌性SD大鼠随机均分为对照组(正常饲料)、低氟组(25 mg/kg)、中氟组(50 mg/kg)及高氟组(100 mg/kg),连续喂养5个月,各组分别于第3、4及5个月处死10只大鼠,分离大鼠下丘脑、垂体和卵巢组织称重,采用氟离子选择电极法测定雌鼠尿氟和骨氟含量,观察氟斑牙的情况,计算各组织的脏器系数;采用免疫组化法测定雌鼠性腺轴组织中TGF-β1表达水平。结果 各染氟组的尿氟、骨氟含量高于对照组(P<0.05),并随染氟剂量的增加和时间延长呈上升趋势(P<0.05);除对照组外,各染氟组均有不同程度的氟斑牙,且检出率随染氟剂量的增加而增加(P<0.05);各染氟组雌性大鼠下丘脑、垂体组织脏器系数于各染氟时间段差异无统计学意义(P>0.05),长期氟暴露条件下高剂量组雌性大鼠卵巢脏器系数高于对照组(P<0.05);长期染毒情况下雌鼠下丘脑组织中TGF-β1表达随剂量增加呈先降后升趋势(P<0.05);在长期染毒情况下各染氟组雌性大鼠垂体、卵巢组织中TGF-β1表达随剂量增加及染毒时间延长呈下调趋势(P<0.05)。结论 燃煤中高氟会影响雌鼠下丘脑、脑垂体及卵巢的脏器系数,会影响雌鼠下丘脑-腺垂体-卵巢轴的功能,其机制可能与高氟影响功能轴中3个脏器组织中TGF-β1表达有关。 Objective To determine the ceofficient variation of hypothalamus,pituitary,and ovarian organsprotein of the models of fluorosis of female rats and the expression of transforming growth factorβ1(TGF-β1)in gonadal axis,and to explore its mechanism.Methods One hundred and twenty female rats were divided into 4 groups:the control group(with common feed),the low fluoride group(content of fluorosis:25 mg/kg),the middle fluoride group(content of fluorosis:50 mg/kg),the high fluoride group(content of fluorosis:100 mg/kg);all the 30 rats in each group were treated with sodium fluoride.The rats were fed continuously for 5 months.Ten female rats of each group were sacrificed in the 3 rd,4 th and 5 th month respectively,and their organ coefficients were calculated.Fluoride ion selective electrode were used to determine the contents of urinary fluoride and bone fluoride of the rats.The hypothalamus,pituitary,and ovarian organsprotein of the rats were weighed,and immunohistochemistry was used to determine the expression of TGF-β1 in the ovaries of female rats while the quantitative method was used for quantitative analysis.Results The contents of urine fluoride and bone fluoride in fluorine-infected groups were significantly higher than those in the control group(P<0.05),and showed an upward trend with the increase of fluoride dose and time.Except for the control group,different degrees of dental fluorosis appeared in the groups exposed to different doses of fluoride,and the detection rate increased with the increase of the dose(P<0.05).There was no significant difference in the organ coefficients of hypothalamus and pituitary in female rats in each exposure group(P>0.05).The ovarian organ coefficient in high-dose groups was higher than that in the control group with long-term exposure(P<0.05).The expression of TGF-β1 in the hypothalamus of each exposure group increased with the change of exposure dose and time(P<0.05).The expression of TGF-β1 in the pituitary and ovarian of female rats were affected to decrease with the change of the dose of exposure and exposure time.Conclusion Fluoride can affect the expression of TGF-β1 in hypothalamus,pituitary and ovary gonads,and mainly affects ovarian tissues.Its mechanism may be related to the expression of TGF-β1 of the 3 organ issues in the gonadal axis.
