摘要
反刍动物瘤胃中酶的活性与瘤胃功能息息相关,为了筛选出合适的瘤胃酶提取方法,实验设计了6种活性蛋白提取方法,分别是超声破碎法、常温研磨法、冷冻研磨法、高压破碎法、反复冻融法和蛋白抽提法。结果表明:超声破碎和高压破碎这2种方法可以提取到总量和活性均高的纤维素酶,高压破碎法可以提取到较高淀粉酶总量的蛋白,常温研磨法可以提取到淀粉酶活性和蛋白酶活性较高的蛋白。说明不同的蛋白提取方法对瘤胃不同酶的总量和活性影响不同。
Enzyme activity is closely related to function in the rumen.To select the optimal extraction protocol of rumen enzyme,6 extraction protocols of active proteins were compared in the study,which were ultrasonication,bead beating,cryomilling,high-pressure press,freeze-thawing,and protein extraction kit.The results showed that ultrasonication and high-pressure press yielded cellulase with highly production and highly activity.Protein with highly production of amylase could be extracted by high-pressure press,and protein with highly amylase activity and highly protease activity could be extracted by bead beating.In addition,different methods of protein extraction had different effects on the production and activity of different rumen enzymes.
