微口膜壳绦虫不同发育阶段及成虫不同节片形态结构的观察

摘要 为观察人工感染小鼠肝胆管中微口膜壳绦虫Hymenolepis microstoma的组织形态结构,以及微口膜壳绦虫似囊尾蚴与虫卵的组织形态结构,本研究解剖受感染的面粉甲虫,收集似囊尾蚴,每只小鼠灌胃新鲜采集的似囊尾蚴50个。感染15 d后,取小鼠肝胆管中微口膜壳绦虫成虫,琼脂糖预包埋后制备石蜡切片,经苏木素-伊红(HE)染色后,采用光学显微镜观察并拍照。解剖微口膜壳绦虫成虫孕节,收集虫卵,使用共聚焦显微镜观察似囊尾蚴与虫卵并拍照。结果显示,在光学显微镜下微口膜壳绦虫成虫头节呈球形,具有4个吸盘和1个顶突;颈节位于头节的后端;未成熟节片内主要分布有肌肉、渗透调节管以及部分未发育成熟的生殖器官;成节内有睾丸、卵巢、卵黄腺、子宫等雌性和雄性生殖器官;孕节内其他生殖器官则逐渐退化,唯有子宫扩大并充满虫卵。共聚焦显微镜下观察到似囊尾蚴呈梨形或葫芦形;虫卵呈圆球形,胚膜内含有1个六钩蚴。本研究明确了微口膜壳绦虫不同发育阶段以及成虫不同节片的组织形态结构,为进一步研究其发育学与病原学奠定基础。 To observe the histological structure of Hymenolepis microstoma in the hepatobiliary duct of artificially infected mice,and to observe the histological structure of cysticercoid and egg of H.microstoma.In this study,the infected flour beetles were dissected and cysticercoids were collected.Each mice was given 50 fresh cysticercoids by gavage.Fifteen days after infection,H.microstoma adults were collected from the hepatobiliary duct of mice,and then pre-embedded in agarose to prepare paraffin sections.After hematoxylin and eosin(HE)staining,the specimens were observed and photographed with light microscope.Dissect the gravid proglottid of H.microstoma adults,collect the eggs,and observe and take photos of the cysticercoids and eggs with confocal microscope.Under the light microscope,the scolex of H.microstoma adult was spherical,with 4 suckers and 1 rostellum.The neck is located at the posterior end of the scolex.The immature proglottid was mainly distributed with muscles,osmoregulatory canals and some immature reproductive organs.There are testis,ovary,vitelline gland,uterus and other female and male reproductive organs in the mature proglottid.Other reproductive organs in the gravid proglottid gradually degenerated,only the uterus expanded and filled with eggs.Under the confocal microscope,cysticercoid was pear-shaped or gourd-shaped.The egg is spherical and contains a oncosphere in the embryo membrane.In conclusion,this study clarified the histomorphology of H.microstoma at different development stages and different proglottids of the adult,laying a foundation for further study on its development and etiology.
