摘要
以雄烯二酮(Androstenedione,AED)为目标化合物,斑马鱼为受试对象,研究不同浓度的AED(0、0.2、2.3和23.7μg·L^(-1))长期暴露60 d后对斑马鱼肠道细胞因子α干扰素(IFN-α)、白细胞介素1α(IL-1α)、白细胞介素1β(IL-1β)、核转录因子κB(NF-κB)和肿瘤坏死因子(TNF-α)的影响。结果表明:与溶剂对照组相比,三个浓度处理组的AED对雄性斑马鱼肠道组织中的细胞因子IFN-α、IL-1α、NF-κB、TNF-α含量没有显著性的影响(P>0.05),但0.2和2.3μg·L^(-1) AED对肠道组织中IL-1β含量具有显著性的抑制作用(P<0.05)。对于雌鱼,AED在2.3和23.7μg·L^(-1)时对肠道细胞因子NF-κB、TNF-α、IFN-α和IL-1α含量都有很显著的提高作用(P<0.05),但AED对IL-1β含量并未有显著的影响(P>0.05)。由此可见,雄激素物质AED对斑马鱼肠道组织的免疫系统和炎性反应有潜在的干扰作用,而且该雄激素对斑马鱼的影响存在两性差异性,即雌性斑马鱼对AED暴露更敏感。该结果为AED对鱼类肠道细胞因子的影响提供了参考,有助于AED对水生生物的毒性评估。
This study was to assess the effects of androstenedione(AED)on intestinal cytokines interferonα(IFN-α),interleukin 1α(IL-1α),interleukin 1β(IL-1β),nuclear factorκB(NF-κB)and tumor necrosis factorα(TNF-α)in adult zebrafish(Danio rerio)when exposed to different concentrations of AED(0,0.2,2.3 and 23.7μg·L^(-1))for 60 days.The results showed that AED at three treatment groups had no significant effects on the levels of IFN-α,IL-1α,NF-κB and TNF-αin the intestine of male zebrafish as compared with the solvent control(P>0.05),but 0.2 and 2.3μg·L^(-1) of AED had a prominent inhibitory effect on the contents of IL-1βin the intestine(P<0.05).For female fish,AED significantly increased the contents of intestinal NF-κB,TNF-α,IFN-αand IL-1αat 2.3 and 23.7μg·L^(-1)(P<0.05),but it did not have a noticeable impact on IL-1βlevel(P>0.05).In conclusion,the androgen AED had a potential interference effect on the immune system and inflammatory response in the intestine of zebrafish,and the effects of the androgen had gender differences in zebrafish,indicating female zebrafish were more sensitive to AED exposure.The results of this study provide a reference for the effects of AED on intestinal cytokines in fish,and help to assess the toxicity of AED on aquatic organisms.
