摘要
目的研究三黄漱口液治疗大鼠牙周炎的生物学机制。方法将60只大鼠随机分成对照组、模型组、三黄漱口液给药组[低剂量组(4.48 g·kg^(-1)·d^(-1)),中剂量组(8.96 g·kg^(-1)·d^(-1))、高剂量组(13.44 g·kg^(-1)·d^(-1))]、甲硝唑组(0.02 g·kg^(-1)·d^(-1))。采用丝线结扎联合10%蔗糖饮水建立牙周炎模型后,每天2次口腔黏膜涂布给药,空白组和模型组给予0.9%NaCl溶液,连续给药1个月后处死各组大鼠,取牙齿样本进行分析。使用ELISA试剂盒检测牙周组织匀浆中白细胞介素-8(IL-8)和白细胞介素-10(IL-10)的水平;Western blot法检测Toll样受体4(TLR4)和核因子-κB(NF-κB)的表达水平;采用HE染色法观察牙周组织病理学改变。结果与模型组比较,三黄漱口液各剂量组牙周炎症状得到明显改善,HE染色显示炎症细胞浸润降低,成纤维细胞增生活跃;牙周组织内TLR4和NF-κB表达降低,IL-8的水平降低,IL-10的水平升高(均P<0.05)。结论三黄漱口液治疗牙周炎的机制与TLR4-NF-κB通路相关,三黄漱口液可降低牙周组织内TLR4、NF-κB和促炎因子IL-8的表达,提高抗炎因子IL-10的水平,改善大鼠牙周炎症状。
Objective To study the mechanism of Sanhuang mouthwash in amiliorating periodontitis in rats.Methods Sixty rats were randomly divided into control group,model group,low-dose Sanhuang mouthwash group(4.48 g·kg^(-1)·d^(-1)),medium-dose Sanhuang mouthwash group(8.96 g·kg^(-1)·d^(-1)),high-dose Sanhuang mouthwash group(13.44 g·kg^(-1)·d^(-1))group,and metronidazole group(0.02 g·kg^(-1)·d^(-1)),with 10 rats in each group.After the periodontitis model was established by silk ligation and 10%sucrose water drinking,the rats were treated with Sanhuang mouthwash or metronidazole through oral mucosal coating twice a day.The control group and model group were given 0.9%NaCl solution.After 1 month of continuous administration,the rats in each group were sacrificed,and the tooth samples were collected for analysis.The levels of interleukin(IL-8)and interleukin-10(IL-10)in the periodontium were determined by ELISA.The expression of Toll-like receptor 4(TLR4)and nuclear factor-κB(NF-κB)was detected by Western blot.The histopathological changes in the periodontium were observed by HE staining.Results Compared with the model group,treatment with Sanhuang mouthwash relieved periodontitis symptoms,reduced inflammatory cell infiltration,promoted fibroblast proliferation,decreased TLR4,NF-κB and IL-8 expression,and increased IL-10 level(P<0.05).Conclusion Sanhuang mouthwash can reduce the expression of TLR4,NF-κB and IL-8 in the periodontium,increase the level of anti-inflammatory factor IL-10,and improve the symptoms of periodontitis in rats.Therefore,the mechanism of action of Sanhuang mouthwash may be related to TLR4-NF-κB pathway.
