PEG-6000模拟干旱胁迫下10种紫花苜蓿萌发期抗旱性比较 被引量:7

Comparison of drought resistance of 10 alfalfa varieties during seed germination period under PEG-6000-simulated drought stress

摘要 为了分析10种紫花苜蓿萌发期的抗旱性,试验采用不同浓度(10%、15%、20%、25%、30%)的聚乙二醇-6000(polyethylene glycol-6000, PEG-6000)溶液模拟干旱胁迫,同时以不加PEG-6000的纯化水为对照(CK),每个浓度3个重复,每个重复50粒种子,发芽期为10 d,第4天计算发芽势,第10天计算发芽率、发芽指数、活力指数、胚根长和胚芽长,利用隶属函数法分别计算10种紫花苜蓿的发芽势、发芽率、发芽指数、活力指数、胚根长及胚芽长6个指标的隶属函数值,再计算各指标隶属函数值平均值,作为抗旱性综合值。结果表明:不同浓度的PEG-6000胁迫对紫花苜蓿种子的萌发具有不同程度的影响。低浓度(10%、15%)PEG-6000胁迫,对部分紫花苜蓿品种种子的发芽率、发芽势、胚根长和胚芽长具有促进作用;高浓度(20%、25%、30%)PEG-6000胁迫,对紫花苜蓿种子萌发具有抑制作用,6个指标测定结果大致呈下降趋势。采用隶属函数法对10种紫花苜蓿萌发期的抗旱性进行综合评价,其抗旱性强弱为WL363HQ>WL168HQ>可汗>WL440HQ>中兰2号>草原3号>三得利>大富翁>佰苜202>敖汉。说明随着PEG-6000浓度的增加,10种紫花苜蓿萌发期的6个指标在干旱胁迫下存在一定的差异,其中WL363HQ和WL168HQ的抗旱性优于其他品种。 In order to analyze the drought resistance of different alfalfa varieties during seed germination period,drought stress with polyethylene glycol-6000 solutions of different concentrations(10%,15%,20%,25%and 30%)was simulated,and distilled water without PEG-6000 was used as the control(CK).There were 3 replicates per concentration and 50 seeds per replicate.The germination period was 10 d.The germination potential was calculated on the fourth day,and the germination rate,germination index,vigor index,radicle length and plumule length were calculated on the tenth day.The membership function values of 6 indexes including germination potential,germination rate,germination index,vigor index,radicle length and plumule length of 10 alfalfa varieties were calculated by the membership function method,and the average membership value of each index was calculated.The results showed that different concentrations of PEG-6000 had different effects on the seed germination of alfalfa.Low concentration of PEG-6000(10%and 15%)had promoting effects on germination rate,germination percentage,radicle length and plumule length of some alfalfa varieties.High concentration of PEG-6000(20%,25%and 30%)had inhibiting effects on seed germination of alfalfa varieties,and the measured results of 6 indexes decreased generally.Based on membership function method,the drought resistance of 10 alfalfa varieties was evaluated.The order of drought resistance was WL363HQ>WL168HQ>Kehan>WL440HQ>Zhonglan No.2>Caoyuan No.3>Sanditi>Millionaire>Baimu202>Aohan.The results indicated that the 6 indexes of 10 alfalfa varieties cultivars during seed germination stage exhibited differences under drought stress with the increase of PEG-6000 concentration,among which the drought resistance of WL363HQ and WL168HQ was better than the other alfalfa varieties.
