摘要
目的基于“肾脑相济”理论,观察艾灸疗法对围绝经期抑郁症大鼠海马神经胶质细胞S100β、CNPase蛋白的影响,探讨艾灸疗法对围绝经期抑郁症的作用机制。方法实验选择40只SD雌性大鼠,使用随机数字表法分为空白组、模型组、西药组及艾灸组,每组10只。围绝经期抑郁症模型由联合去势(OVX)和慢性不可预见性应激法(CUMS)来制备。西药组选择盐酸氯米帕明灌胃,艾灸组选取“百会”“三阴交”和“肾俞”进行治疗。检测大鼠的阴道涂片,采用抓力测试实验进行行为学检测,通过尼氏(Nissl)染色法观察大鼠海马齿状回(DG区)神经细胞的病理变化,采用酶联免疫吸附法(ELISA法)检测大鼠海马S100β和CNPase蛋白含量。结果与模型组比较,艾灸组和西药组大鼠抓力明显升高;大鼠海马DG区神经细胞数量增多,DG区核仁数量、总面积、平均黑度及积分光密度均升高;海马S100β蛋白含量升高,CNPase蛋白含量降低,差异有统计学意义(P<0.05)。结论“肾脑同治”艾灸疗法对围绝经期抑郁症的疗效显著,其疗效机制可能与调控大鼠海马神经胶质细胞的修复与再生有关。
Objective Based on the theory of“Kidney and Brain Complementarity”,observe the effect of moxibustion on S100βand CNPase protein of hippocampal glial cells in perimenopausal depression rats,and explore the mechanisms of moxibustion on perimenopausal depression.Methods SD rats were randomly divided into blank group,model group,medicine group and moxibustion group,and each group had 10 rats.Perimenopausal depression model was established by ovariectomy and chronic unpredictable stress.Results group was treated with clomipramine hydrochloride orally and moxibustion group received moxibustion therapy at Baihui,Sanyinjiao and Shenshu.The behavior was measured by grip test.We detected vaginal pictures,S100βand CNPase protein in the hippocampal by immunoassay.Using Nissl’s staining method to observe the pathological changes of nerve cells in the dentate gyrus(DG)of hippocampal.Results:Compared with the model group,the grip of rats in the moxibustion group and the western medicine group increased significantly;number of nerve cells,total area,integral optical density and average blackness of nucleolus in DG area increased;hippocampus S100βprotein increased,CNPase protein decreased,the difference was statistically significant.Conclusion Moxibustion“Kidney and Brain Simultaneous Treatment”has significant effect on perimenopausal depression,and its mechanism may be related to regulate the repair and regeneration of hippocampal glial cells.
