摘要
目的 :观察肿瘤坏死因子 -α在角膜移植免疫排斥反应中的作用。方法 :将主要组织相容性系统完全不同的近交系F3 44大鼠的角膜植片移植到另一近交系Lou大鼠的角膜上。分为A对照组 ,B同基因移植组 ,C异基因移植组 ,D异基因移植抗体治疗组。治疗组自手术之日起 ,每日结膜下注射TNF单克隆抗体 0 1ml,其他组注入等量生理盐水 ,连续观察 2 8天。对角膜植片透明情况进行评分 ,观察其临床效果 ,利用酶联免疫吸附实验 (ELISA)技术检测不同时间外周血清中TNF -α的水平。结果 :异基因移植大鼠植片平均存活时间为 10 2± 1 94天 ,而抗体治疗组平均为 13 8± 2 17天 ,二者相比差异有显著性 (P <0 0 1)。术后 3天各组大鼠血清TNP -α浓度均有升高 ,术后 7天同基因移植组降至正常水平 ,而异基因组渐趋升高 ,达 95 2±12 5Pg/ml ,异基因治疗组为 78 1± 2 0 5 ,二者相比差异有显著性 (P <0 0 5 )。结论 :TNF -α在角膜移植免疫排斥反应中发挥重要作用 ,应用抗TNF -α单克隆抗体可抑制角膜移植免疫排斥反应 ,延长植片透明时间。
Objective:To investigate the role of tumor necrosis factor-alpha in the rejection of rat penetrating keratoplasty. Methods:44 inbred Lou rats received allo F344 rats or isogenic Lou rats graft,half allograft rats were treated by subconjunctival injection of anti-TNF-α monoclonal antibody (mAb).The serum levels of TNF-α were measured with ELISA at different time points after corneal transplantation. Results:There was significant difference on the serum levels of TNF-α mAb between the therapy group and coutrols(P<0 05).The group receiving anti-TNF-α mAb had a significant delay in corneal rejection with mean survival time of 13 8±2 17 days,comparing with 10 2±1 94 days for the control group (P<0 01). Conclusions:The results suggest that TNF-α plays an important role in the rejection of corneal transplantation,and anti-TNF-α mAb can inhibit the rejection.
