基于Wnt/β-catenin信号通路探讨丙泊酚调控miR-219-5p抑制肝癌细胞增殖、侵袭的作用机制

摘要 目的:探讨丙泊酚调控mi R-219-5p抑制肝癌细胞增殖、侵袭的作用机制。方法:体外培养人肝癌细胞株Huh7,分为空白组、2、5、10μg/ml丙泊酚组和丙泊酚+干扰mi R-219-5p组;PCR检测细胞mi R-219-5p表达;MTT检测丙泊酚对肝癌细胞的增殖抑制作用;Transwell检测细胞侵袭能力;划痕实验检测细胞迁移率;Western blot检测β-catenin、c-Myc蛋白含量。结果:与空白组相比,不同浓度丙泊酚组mi R-219-5p表达升高,细胞增殖被抑制,侵袭能力减弱,迁移率降低,β-catenin、c-Myc蛋白表达降低,且呈浓度依赖性(P<0.05);与丙泊酚各组比较,丙泊酚+干扰mi R-219-5p组增殖抑制能力减弱,侵袭能力增强,迁移率升高,β-catenin、c-Myc蛋白含量升高(P<0.05)。结论:丙泊酚可通过上调mi R-219-5p表达抑制肝癌细胞增殖和侵袭,其机制可能为调控Wnt/β-catenin信号通路。 Objective:To investigate mechanism of propofol regulating miR-219-5p in inhibiting proliferation and invasion of hepatoma cells.Methods:Human hepatocellular carcinoma cell line Huh7 was cultured in vitro,and divided into blank group,2μg/ml,5μg/ml,10μg/ml propofol groups and propofol+interfered with miR-219-5p group.miR-219-5p expression of cells was detected by PCR.MTT assay was used to detect inhibitory effect of propofol on proliferation of hepatoma cells.Cell invasion ability was detected by Transwell.Cell mobility was detected by scratch test.Protein contents ofβ-catenin and c-Myc were detected by Western blot.Results:Compared with blank group,miR-219-5p expression in different concentrations of propofol groups was increased,cell proliferation was inhibited,invasion ability was weakened,mobility was decreased,β-catenin and c-Myc protein expressions were decreased,which in a concentration-dependent manner(P<0.05).Compared with different concentrations of propofol groups,proliferation inhibitory ability of propofol+interfered with miR-219-5p group was weakened,invasion ability was increased,mobility was increased,and contents ofβ-catenin and c-Myc proteins were increased(P<0.05).Conclusion:Propofol can inhibit proliferation and invasion of hepatocellular carcinoma cells by up-regulating expression of miR-219-5p,whose mechanism may be regulation of Wnt/β-catenin signaling pathway.
